As a rabid Mass Effect fan, I've got to say Jennifer Hale. She is Commander Shepard. I've played through all 3 games multiple times as both male and female Shepard, and while Mark Meer is good, it just doesn't beat the kick-ass-ness of some the lines Jennifer Hale delivers.
