    This has already been explained. At the beginning of the game, in Abstergo, when Lucy puts Desmond in the Animus 1.0 and searches for ancestors that Desmond and "Subject 16" share, Ezio appears as a search result. Desmond and "Subject 16" both have Ezio as an ancestor. Hope this helped!
