Depends on the day, I've got at least ten different answers to this one, five of which I use pretty consistently. Mike Tyson's Punch-Out!, Super Mario Bros. 3, Tetris, Mega Man 2, and Tecmo Super Bowl are all viable nominees.

Either Super Mario 3 or Legend of Zelda. Skate or Die is good every once in a while, too.

Apart from the generic "Mega Man, Punch-out, Zelda" answers that are huge classics, I have two other games that are my top favorites:

Super Dodge Ball - classic comedy with good control

Rush n' Attack - Great gameplay and a killer soundtrack

Kirby's Adventure; the first Kirby game where they used the concept that he 'takes enemy powers' and that he was pink. (they didn't do that in the original gameboy games)

River City Ransom: it was a beatumup but you got to beat up Frat Boys and even as a kid I knew that was a good thing.

I'm...so..torn..on what to pick...
I want to say clock tower (even though that was SNES I believe)
and the three adventure games The Uninvited, Deja Vu, and Manic Mansion were so fun....
