Option 1: The war on drugs
While many games do discuss drugs the details of the modern drug culture and the prohibition around them is almost invisible in modern games. Even in GTA the protagonists have a hatred for drugs(Carl Johnson and Niko Bellic specifically). A game that explores how illegalizing drugs empowered organized crime in the 20th century would be something new.
Option 2:Globalization
The world is more connected than ever and this will only continue. The complex nature of globalization(which covers everything from immigration to outsourcing) would make for a very diverse game
Option 3:The pirates of Somalia
Although this has recently been in the news this has actually been around since the 90s. Exploring how fishermen became terrorists would be tense and politically thought provoking
