Just one? That covers an awful lot of room. Kind of like asking "What is the best non-Beatles song of all time?" I really don't think I could narrow it down to just one.
I could easily have listed more. Especially since I specifically included only one song from Andrew Lloyd Webber and Rogers and Hammerstein, to avoid having them dominate the list.

You cannot deny the awesomeness that is... Mr. Feeney as John Adams, going on a tirade and kicking the asses of the incompetent and lazy congress.
I don't know if you can say this is the best musical ever, but it is for damn sure my favorite. This is the musical I grew up with. The humor is fantastic, you can't help but fall in love with the characters. It took me so long to find a DVD of this, but it was worth it to have it in my collection once again.
