It's said in game . . . The plasmids are unstable leading to physical and mental degradation that is only cured by getting more Adam to re-bolster your genetic structure. So it's like withdrawal caused psychosis. (Remember the ghost sequence with the chick in the bathroom, near the beginning, she was trying to cover up the skin falling off her face with make-up).

That said; people flipped out, need for Adam rose, people's pocket books couldn't keep up with their desire, making them resort to the poor house. Frank Fontaine's poor house.

Fontaine tried to rebel against Ryan many times under many aliases. The most recent was Atlas, this is where Jack comes in.

After Fontaine became Atlas he reached out to the people that "Clean the Toilets" and led a working class rebellion against the powerful rich people. This led to Civil War which led to people become more on edge and splicing up more.

Meanwhile Andrew Ryan was abandoning his beliefs to try and stop Atlas and his gang and thus Rapture all fell apart at the seams.

Other contributing factors are:

1)Leakages not being fixed(Rapture Should have been built like a bathtub for it not to sink, Andrew Ryan wanted a city though)

2)The Little Sister program which stole children

I don't get why people are saying the lack of sunlight would cause issues - I've heard of cases where people go made from lack of sun, but I go mad if I'm exposed to it at all.

I think they all went mad with the crushing disappointment that no whales or giant squids or cool things like angler fish ever came by the windows.

I just completed a play-through (on hard with no vita-chambers even!) and I think I can shed some light...

Fontaine shows up and becomes the next big thing in Rapture. He builds a company that winds up bigger and more powerful than Ryan Industries. Eventually, this company, Fontaine Futuristics, invests in the genetic research of one Dr. Tenebaum. She's discovered a sea slug that can secrete stem cells, also known as ADAM. Competition for bigger and better Plasmids breaks out and the heat between Ryan Industries and Fontaine Futuristics only gets worse.

Suddenly, Fontaine winds up dead. Andrew Ryan winds up nationalizing Fontaine Futuristics, which sets angers the populace. Another man, Atlas, takes advantage of this. Eventually, Ryan, who has set himself as governor of Rapture starts wages war on Atlas. The resulting civil war tears Rapture apart. But Ryan winds up briefly the victor. He's gained control of all of the ADAM, which is addicting, and then his Plasmids already have properties that make the people more open to mental suggestion. So, everyone in Rapture who had used ADAM follows Andrew Ryan.

And then that's where you step in.

Put simply, greed. Greed is what killed Rapture.

I think plasmids made people go insane and start to mutate. The splicers in Bioshock 2 look even more mutated and crazed than in the original, due to the passage of time. Also, the way in which Ryan formed Rapture's society (as in belonging to anyone with enough drive to make it their own) probably had something to do with it.
