Any of the bosses in Bayonetta - seriously, every single one of them takes up the entire screen. And might I add that's one of the reasons that game is freaking awesome.

You can't get bigger than Unicron from Transformers (published by Atari, developed by Melbourne House, 2000). Guy's a fuckin' planet transformer for cryin' out loud. You gotta jetpack around him and Cybertron (which he is trying to eat) in the planets orbit and launch your matrix cannon (formed by all the minicons for the soul purpose of combating Cybertron's greatest threat) rounds into his rectum mouth. Which he devours planets with. What?

Bayonetta has the final boss which it's little fingernail is twice the size of you and it's a human shaped god which you have to throw into the sun. Awesome beyond awesomeness. Epic beyond epic.

Calumon: The final boss of Kingdom Hearts... he's big and ugly and he doesn't have shirt and he's haaaarrd! >.<

Probably Elizabeth Greene in PROTOTYPE. She was the same size as a 20 story building but compared to the other answers in this thread, that's quite small.

The biggest size wise was probably the end boss in Bayonetta. The biggest in terms of challenge and epicness was probably Kratos at the end of Tales of Symphonia.

Yep, the sorceror colossus from SotC was the biggest game boss I've seen or killed, unless you count the Over-G akrid in Lost Planet 2, which I don't because it was such a crap fight.

Also, I'm starting to wish there was a Shadow of the Colossus II. That was such a unique and amazing game, though I'd be afraid they'd screw it up if they tried to make a sequel.

I think the largest boss would either be Black Heart at the end of the Ghostrider videogame or the Hive Mind at the end of Dead Space.

Twin-Mold from Legend of Zelda: Majora's Mask was also rather big, but I am not sure if it was bigger or smaller than the firts two. There are probably a few more that I just cannot remember right now, but those are the three bosses I can think of off the top of my head.

Silkroad Onlines Roc.

He's a unique monster and when I stopped playing, he hadn't been killed yet. You actually have to fight other uniques to get to him and he has over 1 billion health.

He appears at about 3:00 in this video.

Second would have to be Medusa.
