With these sorts of things you have to be subtle about how you include it. Don't make a big deal about it. See: Borderlands 2, which makes several mentions to characters in non-hetero relationships (one I remember clearest is Handsome Jack threatening a female worker by going 'You know that wife of yours?' or something along those lines. It isn't picked up on, it's just normal. Weirdly positive effect for a very sinister threat). And of course Borderlands has it's not-so-subtle jabs in the form of the pretty great 'Help the female leader of a town while some **** talks he down for being a woman, then blow up the ****' quest.
But it's the simple stuff, like having a female character dress very much like a man, and NOT correct people when she's called 'Sir', that makes the difference.
