Fancy climbing St Paul's Cathedral? Big Ben? How about taking out Jack the Ripper so he can never murder again? Meeting Queen Victoria? Perhaps even saving her life?

I've always argued that a Roaring 20's America setting would work extremely well. Between the assassins and templars, the mafia also begins to get involved with it's own agenda as they begin to grow in power.
Besides that, picture the outfit. Typical 20's suit and tie combo with a hat and trenchcoat, a hidden blade poking out of the sleeve.
The 20's were a shady era in American history of underground dealings and secrecy, so a suit clad assassin sneaking around and killing high profile targets would work pretty damned well in my opinion.
Don't know if its been posted yet but I don't care Napoleonic era would be fine. Napoleon as a assassin that cracked and went to extremes giving him a whole Fallen hero/He who fights monster motif. and maybe wellington as the assassin Leader.
Edit- And the templar you kill would have to be in the shadows and puppet masters and stuff im sure not that many famous people died at that time for a full game
Oh and maybe youll have to decide who you work for Sympathetic but extreme Napoleon or wellington who could be unsure and cautious and maybe a little ambiguous
Also, perhaps there could be a few missions in attempting (but failing) to keep people alive (Gerhard von Scharnhorst, Horatio Nelson, Prinz Ludwig-Ferdinand von Preussen...
I'm not certain about my history of the Napoleonic Wars (something that must be fixed!!) but wasn't Nelson killed by a musketball? Could be an interesting race against time to kill the sniper...

Ah yes, I have played that game. Only for a few hours, and not very much (prefer the openness of Empire personally) but it did give me an interest in Napoleon. Should really install that game again. There was nothing really wrong with it.
That would be a very interesting mission. Assassin's creed during the Battle of Trafalgar. I mean, the trailer for Revelations does show ship-to-ship combat.

No, you're quite right, he was shot from the mizzen-mast platform (don't know the naval nomenclature of those things) of the Redoubtable by a French marine. He died about four hours later, so hooray for another cutscene idea!
I think that mission would be along the lines of manning guns for the first bit (taking out the Bucentaure or other lead French ships), then boarding another ship, capturing a colours while clobbering Frenchmen (sorry as an Englishman, I found a perverse pleasure in typing that!), then noticing a marksman line his sights against the admiral finding a musket, aiming... only for it to misfire (possible and very plausible, if not probable), hence Nelson gets hit. Then finding some other way of killing the marksman and returning to the Victory.
That would be so incredibly cool. Brotherhood's final mission, the siege, was pretty epic, but being at the Battle of Trafalgar would be so much more so...

At the end of Brotherhood, when they found the Apple, Desmond used it to ask where the Temples are located. Shaun, upon seeing and identifying the Phrygian Cap (which stands for freedom) and the Eye of Providence, said "both come together in only one place." Though I can't remember if it was explicitly mentioned in game, that "one place" would be "The Declaration of the Rights of Man and of the Citizen," which was adopted in the late 1700's in France during the latter days of the French Revolution, and was a precursor to France's Constitution after their revolution.
So, it seems to me that ACIII would likely take place in France, with Desmond exploring modern France in search of (censored for possible spoiler). Though it also seems possible, and likely, that Desmond may use the Animus to relive the memories of a French ancestor Assassin who operated during the French Revolution, and these explorations in the past would help him navigate modern France, introducing a strong dual-gameplay component between Desmond and this yet-to-be-named ancestor, which, to me, sounds awesome.
We've had just a hint of this dual-gameplay between past and modern in ACII and Brotherhood, and I would love to see how it could play out if they had Desmond and his ancestor exploring the same areas, but in different time periods.
EDIT: As a real-world history tie-in, they could use the legend of the Scarlet Pimpernel, possible as an ally, and say that the Templars had infiltrated the royalty and aristocracy that the people were rebelling against, which makes sense because it has been mentioned in Glyphs in Brotherhood that, for the longest time, the Templars controlled the world through monarachy, and as modern democracy spread, they had to adapt their methods. Also, it could show the potential ugly side of taking down the Templars: many people who didn't deserve to be killed were beheaded by the revolutionaries for no reason other than the fact that they had money. This would highlight the morally grey status that the Assassins occupy: yes, taking the Templars out of power is always good, but what about the other extreme, knocking someone off because they MIGHT be a Templar? So interesting...

Actually, I was thinking that a final mission at the Battle of Waterloo would be even more epic, in and around La Haye Sainte, and trying to kill Napoleon (but he escapes), all the while fighting off waves of Old Guard (equivalent of heavies). It's got everything: artillery fire, cavalry charges (both being in one and fighting one off), plenty of CC combat, wallrunning around the hamlets etc.
Final cutscene would be on the HMS Bellerophon at last capturing the Emperor. That'd be kinda cool, I think.
The last boss fight could be a tag time fight where you and Gorbachev fight Ronald Reagan and Margaret Thatcher on top of the Berlin wall while it's being torn down.
The idea's not that far off, though. A lot of the material that came up in the Subject 16 puzzles in Brotherhood was very pro-Communist, anti-Capitalist, so it's very easy to see the Assassin Order being behind Socialism/Communism.
Personally, I want to see modern-era Desmond using his newly developed Assassin skills. I know the temptation to stick guns into it might exist, but I think it would be awesome if some crisis befell the Assassins, cutting Desmond off from his support lines, and forcing him to run around a modern day city, using his runny-climby-jumpy-sneaky-stabby skills against gun-toting enemies. If it was designed and paced right, it would be a very tactical and smart game, forcing the player to play stealthy and smart against more powerful adversaries.
I'd like to see ACIII set in the Aztec capital, Tenochtitlan. It's supposed to have been an amazing city and the Aztec culture is fascinating, but I'm not aware of it having shown up in a game before. If the story was set around the the time of the Spanish conquest it might even be possible for Ezio to put in an appearance.
Actually an AC set in Japan would be freakin' awesome... Though maybe it could act like a completely different division of assassins from those in the West, with different rules and creeds, with only a loose resemblances? Perhaps we could be exploring the memories of a new protagonist that would allow for them to use authentic Japanese fighting styles of the shinobi, rather than having the 'hoods and hidden blades' of previous AC games.
After having a long talk with a friend of mine, I realized just how viable a game set in Feudal Japan would be. Now, the argument I hear a lot is that Christianity didn't exist in Japan during the Feudal period, which... is vastly incorrect. I suspect, if gone this route, the game would take place in the year 1637, during Shimabara Rebellion.
Christianity had begun to spread like wildfire throughout Japan's peasantry. The shogunate suspected that the Christians no longer had loyalty to their daimyō. The government began persecuting Christians. Thirty thousand Christians, ronin, and peasants rose up in rebellion, forcing the samurai to put them down.
Now, think of it like this. Japan is known for several things when it comes to 'badassery', one of the primary things being, of course, the Shinobi, or the man of shadow. What if Japan was a nation controlled by the Assassins? What if the Christian movement was an attempt of the templars to try to unseat them? You could also do the direct inverse. Assassins also seem to be Christian, more or less. What if the samurai class are templars? Discuss, and feel free to bring up other locations and arguments.
(And no, don't bring up France, or WWII. Ubisoft basically outright said no.)
Honestly I see them most likely using the character from there comics who is russian and I believe it takes place during the cold war. There is also alot of references to the cold war throught the games so I think its a pritty fair assumption.
That would be the first ever justified human boss character, it would be interesting coming into a battle that in order to be historically accurate has to have the opponent surviving multiple bullets and stabbings, as well as whatever else you can throw at him.
What about Egypt at the time of the Victorians (1880s-ish) (When England was in charge of it) Cairo would have plenty of freerunning potential, and there was the whole Ancient Egypt tomb robbing craze going on, so plenty of mysterious ancient artifacts being dug up and needing to be retrieved. It could be like a spy game, as you and other assassins try to find out where the best digs are happening, while the Templars are doing exactly the same thing, but you have to be discreet, and kill anyone who discovers your identity or gets in your way. You could even be responsible for assassinating everyone involved in discovering and opening the Tutunkamen tomb! What a plot twist.
