I think the moment I stepped out of the escape pod in Halo: Combat Evolved, that was the moment when I knew I was destined to like that game.

Also, it was the moment the thirties music and atmosphere came up in Bioshock that I knew I would be entranced.

Finally, it was the combination of the Ashley/Shepard romance scene and the Illos/citadel mission with Wrex and Garrus and the general feeling of epicness that made Mass Effect stop merely being a 'damn good game' to a 'oh hell yeah this is one hell of an awesome game' sort of feel for me.

Lets see Jet Grind Radio at first I liked it a lot but I was a kid I couldn't appreciate it. Then I played it when I became older and I love everything about that game. Also MGS I should have known I loved it from the countless times I played the demo. Then I beat it three times on Playstation, Once on PC and using an emulator, and twice on the GC. I'll probably buy it from PSN and beat it again lol.

Freedom Fighters- Storming Ellis Island in a scene like D-Day.
Star Wars Republic Commando- Final missons of act 2 where you must defend the ship from..... hell that whole act was awsome.
Mass Effect- So many scenes, but the final decision was one of the most epic ever.
Ratchet and Clank Up Your Arsenal- The "My daughter tells me your good with your hands." scene.
Knights of the Old Republic- Learning of your past.
Half-Life 2- Attacking Nova Prospect with an antlion army.
Halo- Level 9 is a free for all between Flood, Covenant, and you.... guess who wins.
Gears of War- Beginning of Act 3 is full of rain, wretches, and immulsion.
Mass Effect 2- Recruiting Archangel, later on recruting Tali, plus Tali's loyalty mission are all crowning moments of awsome! Plus punching out the reporter and letting Harkin take the bullet to the kneecap just for shits and giggles, then reloading cause your paragon :(
Gears of War 2- Act 3 is full of rain, wretches, and pissed off science experiments. (What can I say, I'm a sucker for the rain affects in Gears of War)
Elder Scrolls 4 Oblivion- Moment I got out of the tutorial dungeon.

Thats all I can think of at the moment.

Stepping out of the Vault in F3
Stepping out of the sewers in Oblivion
Standing at the fortress of the Blades in Oblivion and looking down at everything.

Jumping into the portal after Marle in Chrono trigger. "for the first time"

The moment I realized what one of the side missions in Saints Row 2 was, you were slinging poo at people and getting points for it! Also, the very first mission where you bust into a courthouse and slaughter everyone to get one guy out and calmly walk out talking to him as if it's no big deal.

Persona 3: Watching the intro cutscene and immediatly drawing parallels to Evangelion
The Worlds Ends with You: Neku's opening monologue "I don't get people, never have, never will."

Kingdom Hearts, the moment I got to The Nightmare Before Christmas world. I liked the game before, but that step into my childhood finalized the deal and it's been love for the series ever since.

Team Fortress 2, my first 28 kill round, it just made me feel as if I had overcome the console handicap.

Deadrising, the minute I ranked in the top 200, I realized that this was a game I had put far too much time into. But also, the lawnmower, or the sawblades...

In Xenogears the part that really got me was...

At end of disc 1 when Weltall blasts up through the floor behind Fei while he changes to Id and destroys Solaris.

Fucking epic.

I didn't see any sort of connection between evangelion and p3.

other than metroid Prime

Kingdom Hearts the opening video was amazing to me and I loved it right then and there.
When Riku smiles on the other side of the doors to kingdom hearts as they seal. I am not going to cry!

I fell in love with Metal Gear Solid at the helicopter pad

Orcorina of Time, must I say more?

In GTA 4 when you are doing missions with Little Jacob. Then when you steal the helicopter with him and he says that the two of you should just take a the chopper and fly to Jamrock. Then when you have to decide between your cousin and the girl.

This. I had never seen anything like it at the time, and it's still my favorite game in the series. This was the game that got me back into gaming. And it all started with this scene.

Also, it reignited my love of orchestrated music.

Emerging from the Vault in Fallout 3 is definitely going to be mentioned several times, I think. They did a great job capturing that "beautiful; but what now?" feeling, assuming that has a proper word for it.
I tend to agree with Yahtzee that the effect was blunted a bit by being a bit too reminiscent of Oblivion. Coming out of Vault 101 was a bit like the second time I had sex. I'm sure it was good, but it wasn't particularly memorable by comparison to the first time.

On-topic, when I got a Roman general in Rome: Total War fresh out of the loony bin leading my troops...and I listened to some of his speeches. To this day I still let the generals talk, and after over five years it still hasn't gotten old.

Okami - I found the dialogue hilarious and the art was awesome
Assassin's Creed 1&2 - When I felt like a total badass assassinating someone and escaping
Halo Wars - When it reminded me of my awesome childhood playing Age of Empires
Portal - ..... Do I actually have to explain this one?
Left 4 Dead 1&2 - I just like the way you have to act as a team, it's no where near as fun when you play it by yourself..

O.O Four pages of replies. In about a two days. I'm very impressed...and pleased.

I left out a few good games I fell in love with, so I'll list them here:

Tomb Raider Legend: When I did a swan dive off the falls in Ghana.

Batman Arkham Asylum: When I heard Kevin Conroy and Mark Hamill as their respective roles once more.

Final Fantasy X: When I heard the Hymn of the Fayth for the first time.
