Ahh so comedy groups that do music...I was thinking more Bad News, Boyzvoice, or CB4. I'll post some vids of those because they are hilarious, they are in the Spinal Tap vein.

Bad News, the ultimate metal losers. This song used to get covered by every band in the town I was born.

Boyzvoice from the Norwegian film "Get Ready To Be Boyzvoiced".

The great CB4...I think he may be black...

As for comedy groups that do music...well my heart belongs to DAAS but Tripod can have my money.

Doug Anthony All Stars. I was very impressionable at the the time and they pressed hard.

Tripod. Best group of its kind.
For more D&D track down Tripod versus the Dragon.

Not the greatest but still enjoyable, Tripod:
I couldn't find any good clips, but I also like The Hot Dog Man, Maryanne and On Behalf Of All The Geeks.

Besides some of those already listed (ie Stephen Lynch), I'm a fan of Bloodhound Gang.

Possibly their best known song, Bad Touch (aka Discovery Channel). Warning for lots of sexual innuendo.

Uhn Tiss is my favorite song though. Warning: Honestly, it probably doesn't even need a warning, but there is implied sexual contact. It's much less risque than a Mass Effect sex scene and is the punchline of a joke they take the whole video to set up.

Their other songs I enjoy:
Lift your Head Up High(And Blow Your Brains Out)
I Hope You Die
No Hard Feelings
Right Turn Kyle (These last four are awesome anger/hate songs, especially No Hard Feelings for a break up)
Three Point One Four
I'm The Least You Could Do
I Wish I Were Queer So I Could Get Chicks
Mope
The Ballad of Chasey Lain
A Lap Dance Is So Much Better When the Stripper Is Crying

I'll just add nothing new and say Bo Burnham, Tim Minchin, Stephen Lynch, and Jonathan Coultan. I don't know how to put in videos, so yeah... If mrweebl counts then him too.

Psychostick is very good. Songs like Because Boobs, Do you want a Taco?, Sadface:(.

Love it all
This. A thousand times this.

Also Jon Lajoie, a Canadian comedian who does some music. (not really a band but whatever)

Not sure I have a favorite, though Weird Al probably has seniority in my mind. I haven't listened to more than one or two songs by a lot of the bands already mentioned (Coulton, Bloodhound Gang, Stephen Lynch...). I even have kind of a lingering liking for Allan Sherman songs. Tenacious D is always awesome. And does Parry Grip count?

Still, Psychostick is who I've been loving recently.

I liked it when they just went ahead and <url=http://www.youtube.com/watch?v=uORxfNQ0qyE>recorded a sandwich.

So good people tend to forget they began as a TV joke skit:

If you mean bands that compose their own music however I've always been an Arrogant Worms fan myself.
