I'm going to have to go with Tess from the Jak and Daxter series. She's not really in a leading role, but she falls in love with a little furry rat... And then turns into one. Now that's what I call an interesting love-story.

The problem I have with female Shepard is that the game was so obviously designed for Shepard to be a male. Well, not obviously, but you can still see she's tacked on. She never shows up on box art, she never shows up in trailers etc. They clearly had a man in mind when they designed her so to speak.

Don't get me wrong though. I love female Shepard (in the sane "I love the fact that she' in the game" kind of way), It's just a bit sad that all we see of Shepard is the stereotypical bald male white space marine.

My personal choice is Faith from Mirror's Edge. Just because her sex appeal was deliberately downplayed. She's not even that pretty, not intentionally ugly, just not hot. Is she a deep, interesting character? No, but she stands against everything that is wrong with female leads in video games.

This just reminded me of a conversation my mates had ages ago. I think overall we decided Meryl from Metal Gear Solid and Alyx from Half-Life 2 were two of the best.

Jen from Primal and Zoe from Dreamfall. It's not often you can make a girly-girl the protagonist in a game and have it work.

Though I have never played it, I hear Jade from Beyond Good & Evil is pretty cool.

Also, Ellen from Folklore. I believe she deserves special mention for being a female protagonist in a Japanese-produced game who does NOT have the emotional fortitude of moist tissue paper.

i wouldn't say the best, but lara croft is probably one of the most memorable, due to the tomb raider games/films being everywhere for many years. i don't know if i could say best for definate, but samus would be the obvious choice.
