Defense Grid, no doubt.. It fairly hard, especially the Aperture Science maps..

Defense Grid with the DLC is my personal favorite. Tons a map types and difficulties to keep you coming back to the game.

The most fun to pick up and play would be Plants vs Zombies. The only thing keeping that one from being the best is that it's actually a pretty easy tower defense game. More for the casual gamer, but extremely fun to go through just the same.

Dungeon Defenders is good, but if you play co-op online you can't just pause the game and leave. I haven't played local co-op so I don't know you can pause that. It's funner and easier with other people than alone. If you want a game that you can pause and put away whenever you want I don't think Dungeon Defenders is a good choice.
Orcs Must Die however should be fine, so I'd recommend at least trying the demo to see if you like it.

thanks for the replies, folks. I Orcs looks interesting, I may start there. it looks a little silly for me, but I like the idea that you can participate in the action in addition to "placing the towers".

One other thing, I have Starcraft 2, but I never play it. Are there any good tower defense maps/campaigns there?

For SC2 Custom games:

Element Tower Defense! I like that one a lot.

Roundabout War: is something of an inverse TD thing (you make buildings to produce units who go kill guys in the middle) I'd also recommend at least trying that. It's so much fun to watch all the massive amount of destruction going on in the middle.

There are plenty more, but I don't do customs a whole lot.

I'd cast my vote for Dungeon Defenders, but it's barely a tower defense game at all, at least in the traditional sense.

Came across thi recently, difficulty curve is quite smooth and the basic formula is simple but allows for a viriety of tactics. Also it's quite tongue-in-cheek.

Also, plants vs. zombies, which I count as TD because I can't think of another genre into which it fits.

Gem Craft is a really fun flash TD, and it's the only one I've really played for a very long period of time.
