My favorite let's play is probably the ones done by the Aussie Gamer, particular the Hitman: Blood Money ones. Man those are classic. I like how he includes the screw ups and oddities in the videos.

My absolute favorite and one I watch quite often is Matt and Woolie's Old School Playthrough of Donkey Kong Country. Every part is entertaining.
Close to that is Matt and Pat's Resident Evil 2 playthrough, mostly because Resident Evil 2 is one of my favorite games as well.
Kind of wish I had editing software and a good microphone and the attention span to do lets plays. Aw well.

One of my favourite LPs is the Mass Effect LP by The Freelance Astronauts (SAMaxwellAdams on youtube). Not only are these guys hilarious--and their other LPs are well, well worth watching--they completely break the game in every way they can. Here's one of the funniest samples from that LP:
The best Lets Plays is MackAndMesh, they play scary games exclusively and Mesh is a real scardy cat. Probably the first time I've ever laughed so much seeing someone get so scared! not to mention they where the first ever to start doing Lets Plays, before youtube came along.
