Everyone has a sequence in a game where they always enjoy it, no matter how many times they play it. Now this sequence may be a quest, dungeon, boss, cut scene or even a musical score. Some people enjoy them for their challenge, or their beauty, or even because it makes them think. It may help them connect with the characters, or even help define their own character.

Here are some of mine.

Dungeon/Level: The Ludo Kressh level from KotOR II. It made you realize that the Exile was a character who had a life before the game, and helps shape his morality.

Same with Moridin's loyalty mission from Mass Effect 2. It made him reflect on his actions and how the krogan are a people, not just a statistic. It was one of the few times he didn't seem to have a correct answer about anything.

Cut Scene: Shadow Hearts: Covenant is probably my favourite and most memorable game since it was one of the first games a friend and I marathoned at sleepovers. It blew our 12 year old minds away. Its not the prettiest game, and the only video I could find was filmed with a video camera, but here's the opening of the game.

Also, to completely ruin this game's image of being serious here's the comedic relief character.

Leon you so cool.

Edit: I realised I could have placed something intellectual or thought provoking here but instead I went for backflips and slo-mo.

The ending of Assassin's Creed: Brotherhood really surprised me. I don't want to spoil it, but I'll say that the way they subtly took control away from you (gameplay transitions seamlessly, and not all at once, into a cutscene) had me engaged in such a way that I would not have expected from a game that isn't Heavy Rain.

The 'Death From Above' mission on COD4 is one of my all time favorite levels. I've probably played it more times than any other level from any other Call Of Duty game.

And in Oblivion, my favorite mission is the Dark Brotherhood contract 'The Night Mother's Child' where you stalk the party goers and murder them one by one.

The final final boss from Ocarina of Time. Hell, both final bosses were great. I know some people are partial to Ganondorf, but that last battle was just so epic and atmospheric.

Also, strangely, the beginning of that one mission from MW2 where you start in a bunker and walk outside to see the burning D.C. at night.

I think it's been out long enough that I don't have to tag this for spoilers so here it goes...
My favorite sequence in any game has to be the final boss fight in Just Cause 2. Hookshotting yourself back and forth between 4 launched ICBM's while shotgunning down a Kim Jong Il stand in has to be the most ridiculously awesome thing I've ever done in a game.

1. Beginning of Bioshock.
2. Lair of the Shadow Broker
3. Opening to Arkham City. (I'm not too far in yet, so I expect there to be other moments).
4. My full memory of it is very fuzzy, but I definitely recall loving the space sequence in FF8.

Is that what the quest is called in PAL regions or something? It's one of my favourite game sequences too, but here it's called "Whodunit". Your title is a good sight better.

My own choice would probably be the water section of Amnesia: The Dark Descent, or perhaps any given cutscene from Bully.

And THAT scene from Heavy Rain as well...anyone who's played it will know which one. It's pretty hard to watch.

Take it Back! in Fallout 3,

Storming the Jefferson Memorial (I think) with a hugeass robot blowing everything up, having tons of Brotherhood of Steel knights fighting for you, plowing straight through Enclave checkpoints and blowing everything to kingdom come.

Afterwords in the building is still fun and satisfying to kill the Colonel, but it lacks explosions and commie-hating mechas.

That's what the quest is named/called on the Oblivion wiki site, or at least that's what the journal entry is titled. I just prefer Night Mother's Child over Whodunit.

I assume you're talking about how Desmond kills Lucy correct? My problem with that is that you still have to use the controller to actually force him to kill her. As in she doesn't die without your input, therefore contadicting the entire "you have no control" vibe the game was going for.

I think a trying to hold yourself back QTE would have worked better, the difficulty of which would have to quickly escalate to insane mode (think Heavy Rain on Insane Mode)

As for myself... Two words, Myriad Truths

EDIT: In hindsight, Silent Hill 1-4 also

No one mentioned Dead Space 2 yet?
Say what you want about that game, but it most certainly had some awesome gameplay sequences...

Resident Evil 4 - The cabin sequence where you keep zombies out while being forced up the stairs.

Final Fantasy 6 - The Opera House.

Dragon Age Origins - The Landsmeet.

Dragon Age 2 - The Arishok confrontation.

This is going to make me sound like a PS3 fanboy, but:

The Bertrand chase sequence in inFAMOUS 2

The horse riding sequence in Uncharted 3 (I don't want to spoil it, but for those of you who've played it, you know what I'm talking about)

-While Arkham City improves on the original, it has nothing to top Scarecrow's sequences in Arkham Asylum, his intro is one of my all time favourite wtf moments in gaming.
-The Strider finale in HL Ep 2
-The first village scene in Resident Evil 4. Set the tone beautifully, stressful without being unfair
