The "I am not left handed" line would be one of the best, just because it takes a few seconds to actually work out what they mean.

slightly off topic, but I'm certain this exchange happens every single time someone new is asked to watch it:

"Hey, wanna watch a movie?"
"Sure what is it?"
"It's really good, it's called the Princess Bride."
"Pfft, I'm not watching a pansy movie like that" *30 minutes later* "This is the best movie EVER!"

I always loved its two most famous scenes: The sword fight between Inigo and Westley, and the poison trick scene. Every part with Wallace Shawn or Mandy Patinkin is awesome, though.
