Thinking about it i came to a stunning conclusion.
Since i never really played (as in: i never finished them) any adventure games, moneky island, day of the tentacle, sam and max etc. are out.
I don't think Portal was that funny to be honest. I had a few good laughs, but other than that it seemed far too serious. Same goes to a lesser extent for Portal 2.In addition i hate Wheatly with a passion.
Serious Sam has funny gameplay and monsters but isn't really that funny in itself.
Why people consider Borderlands funny is completely beyond me, i can't remember anything funny about that game. Maybe i just have a shitty memory (or sense of humor).
Dwarf fortress is really funny at times and while this is most definitely part of the game design the humor always feels a bit detached from the game. I really don't know how to explain it, sorry.
So in the end i might actually have to go with DA2. The Companion dialogues were in my opinion the best thing about the game.

I'm actually pretty sure that i would be able to think of a better game, but right now nothing comes to mind.

Portal games for dark humor, Saints Row games for immature madness, Earthworm Jim for complete randomness, Katamari games for that special flavor of Japanese crazy.

Like many people on here, I'm probably going to go with Conkers Bad Fur Day as all-time funniest.

This gen, I'd have to go with Shadows of the Damned. Any game capable of squeezing that much innuendo into one campaign needs the recognition.

Portal 2. Portal too, but it's humor was more downplayed. And there's more stuff in the sequel.

Also Ace Attorney series and Ghost Trick.

Borderlands 2 was hilarious, but not as funny as Ben there, Dan that, a download-for-free indie adventure game. It's so freaking brilliantly written.

I'm really not sure what I consider the funniest. I don't think Deadpool even ranks in my top 10 as it tries way too hard. Most games completely forgo any form of subtlety anymore though so I'll forgive anyone who thinks otherwise.

Conker's Bad Fur Day: Great game, fun N64 title that kept me in stitches from start to end, even playing through it now I laugh insanely. The concept is you play as a rude, foul mouthed squirrel with a hangover on his way to save the day.

Bard's Tale: Some of the best humor in a game to date. The game uses some subtle jokes and some overt ones, but there is comedy to be found in every step of the game if you look for it.

Blood Dragon: Not very subtle, but that is intentional. It is a complete reference point for the late 80s early 90s movies. If you grew up in that era, this game is for you. Even if you aren't that into shooters (such as my girlfriend) you will end up falling in love with the game. Unfortunately the game itself is pretty bland.

The Monkey Island games hands-down. Nothing can come close in terms of pure comedy genius. My particular favourite from the series, was Murray the Demonic Skull. The fight in the first game with the Sheriff in the mansion was also legendary and still stands to this day as one of the funniest moments in gaming of all time.

I'd probably give Portal a second place. Glados was very funny. Shame there's not more humour in games.

I'm genuinely kinda surprised no-one has said this, but: Ratchet and Clank
all the ps2 and the ps3 ones up to "a crack in time" are pretty funny, although the funniest is probably 3 (or "up your arsenal" for Americans)

after that...well, Tales of Graces f has some fantastic moments, especially in its skits (I'd say its quite a bit funnier than blazblue even, so if you liked that....)

for actual gameplay, its gotta be Just cause 2/red faction guerrilla, or possibly split/second.

seriously, split/second is the funniest game in the world if played with a group of friends in the room (ideally whilst a little tipsy, but there it is)

Jak 3's act 3 plot twist (you know the one) will forever be one of my most historical moments playing a game. I absolutely lost my mind, reeling in laughter for far too long after that.

For funniest game in general, It'd either have to be Portal 2, Persona 4, or... anything from the Ratchet and Clank series.

The old Sierra and Lucas Arts point and clicks had great humour. Sierra games in particular had some very quirky and often biting humour about your actions.

The most humorous recent game I say I've played would have to be Portal 2.
