Portal and Sam and Max. Portal is pitch-black deadpan comedy gold, and Sam and Max is funny because of the absurdity.

Not really funny because of the content but Halo 3 is funny on forge (especially when playing rocket launcher baseball with a grav hammer).
Messing around in SSBB was also fun, I remember playing one time with smash balls as the only item. It was frantic.

No more heroes had some funny moments.

Space Quest IV: Roger Wilco and the Time Rippers, especially with the time trip to Space Quest I and Space Quest X: Latex Babes of Estros.

I'd have to say that the only two sort of funny games I've ever played were Saints Row 2 and The Matrix: Path of Neo.

Saints Row 2 was genuinely funny. Not bent-double, gasping for breath funny. But occasional-chuckle funny.

The Matrix: Path of Neo, however, was not genuinely funny. And only brought about laughter of disbelief when I was fighting giant, bipedal ants in some kind of pan-dimensional hubworld where everything is either chequered or a door and some things catch fire for no reason.

Also, it was a really shit game. But the cutscene before the Giant Smith boss put a smile on my face.

I'm gonna go with Ratchet and Clank 3 on this one. Yes, the humour is childish, but I don't care, it still had me buckling at every line!

As for funny things happening... Halo 3 and SR2. Whether you're blowing up car after car or creatively arranging grunts to fly off team-rocket style (Yahtzee) in the most spectacular way possible, these two are the most fun you'll have with a gun and a sword ever.

Lego Star Wars wins my vote. Probably the first time I've ever laughed at a moment that the developers had intended to be funny.

Im going to have to plain out and say Earth Defense Force 2017 is easily one of the pinnacles of game humor. It succeeds in combining brilliant, witty camp with HEAVY satire of almost every shooter out there (and action film). AND it surprisingly has fantastically enjoyable gameplay.
In a close second comes Dungeon Explorer! for the combination of Five Player MP (Yes Five) and the hilarious, and occasionally subtle, parody of rpgs in that era (even some of the gameplay elements are satirical), even while having some of the most insanely difficult bosses ever.
Reeling in at third is Psychonauts, and if you've been reading the thread you've probably figured out why.

Honorable mentions:
TF2, Conker's Bad Fur Day, Black and White, Duke Nukem, And all you crazy big budget games that don't realize how ridiculous you are.

For all you people who include things like Halo 3, and SSBB, please realize that literally everything in existence can have situational comedy, only intentional comedy is actual comedy.

You guys have mentioned just about any funny game I could think of off the top of my head...EXCEPT!

Earthworm Jim games.
MDK games.

Guns and ammo, strange terrains, hilarity ensues.

Add Conkers BFD to my list of hilarious games, just realised I missed that one.

Oh, and Dynamite Headdy and Chuck De-cap were both pretty amusing ideas. I mean, a puppet who throws his head at people, and a mummy who punches people with his head... located in his stomach!

Conker's Bad Fur Day would have to be my pick for all time favourite. The humour wasn't subtle at all but it was hilarious to me even so. How many boss fights involve flushing a giant poo after it sings a song?

Also Majesty. That game had a underrated sense of humour. The characters had some funny lines, you had to bribe them to do anything you wanted and the cheats were funny to put in and the confirmation messages often made me chuckle

The Meccaryns from Giants: Citizen Kabuto are some funny characters. Too bad it wasn't enough to make up for the horrible marketing this game got, as well as the crippling bugs in the engine. (hi2u Desktop for no good reason)

It definitely had its moments though. Here's a clip, the Meccas are those dudes in the suits.
