I love rts and city builder games, and tbh with mothers day around the corner, I'd love this to be the year I got my mom into gaming.
I'd probably recommend the Sims and a Tycoon game to get her off the ground. My mum gets pretty nervous about games where you can get killed by enemies, even with stuff like AoE she gets kind of upset when armies come and knock down her hard work.
She's probably most obsessed with the Sims, I guess if your mother's a motherly kind of mother she might appreciate the whole looking-after-people-and-building-a-family aspect of it. Or, of course, she might want to just get away from that :p so I guess it depends on your mum, but most people's mums I know love the Sims if they like games at all.

I know someone whose mum's obsessed with Final Fantasy, actually. She's a Reverend.

I've already tried portal, but she didn't know what a portal was, and also how to move in an FPS. We're not ready for that yet.

Point and clicks may be a good first start:

Broken Sword and Monkey Island games are quite good with some humor (especiall MI) and not all that hard to play, avoid kings quest though, not a good one for a first timer.

Yeah, this. My mom spent months trying to convince me that I wanted a Wii, just so she could use it. Never mind that I don't even live at home anymore.

Anyway, I'd say something casual and Wii-esque is your best bet to get her into games.
