Gonna state the obvious here: Sheogorath. Shivering Isles is still my all time favorite expansion ever, simply because of his dialog.

Henpecked Hou from Jade Empire is also one of my favorites.

 

[Soxafloppin]

Zeke from inFAMOUS always entertains, particularly in II. Sully from Uncharted is great too, I love his authoritative, disproving, older man-ness

 

[putowtin]

The Boss from Saint's Row 2 onwards. Somewhat dependent on which voice option you pick, but there's a lot of really funny lines.

Tru Dat!
Go with male voice 1 (the cockney) and it's hours of fun! (just replaying the series for the new one)

Deadpool from the recent Deadpool game gets a mention as well, if just for his zany antics as opposed to actual "humor."

Pretty much half of the cast in the Paper Mario games. From the likes of Mario's party members, to the likes of Professor Colorado, or even Bowser of all people(or koopas I should say). Just about every character will say something chuckle worthy sooner or later.

Seeing what happened when you tried to attack him was probably the funniest Elder Scroll moment I've had.

Junpei from Persona 3 tends to get into all kinds of funny situations.
The way Yukari always brings him down is hilarious.
Or when he took a flashlight out of nowhere and started telling ghost stories! "Welcome to Junpei's Believe It, Or Don't"
And he's definitely a wealth of funny quotes
"He's not gonna make us walk the plank, is he?" when he first met Mitsuru's father
"Tartarus? Sounds like a toothpaste"
