What do you like?

What is funny is very much opinion, so can't really guarantee you'll find something funny without knowing what your sense of humour is.

Anime- I find Panty and Stocking with Garterbelt to be hilarious but I suppose it's an acquired taste, for a lighter example by the same company try Hanamaru kindergarten (hanamaru youchien)
Tv Show-Community was very surprising and fun. I'm glad that smart nerd-comedy can still be popular.
movie-I will always say Young Frankenstein was the best comedy ever made bar-none.

Finally saw Clerks 2 and it was pretty damn funny,so is Anchorman. The Intern arc on season four of House is hilarious due to House basically just fucking with the new guys.

Also Sayonara Zetsubo Sensei is funny though the jokes are a little more Japanesey if you know what I mean.

If you like dark comedies, Death to Smoochy, 8 Heads in a duffle bag, Slither, Shaun of the Dead and Bad Santa are all pretty funny.
If you like non-dark comedies, 'How to succeed in Business without really trying', Galaxy Quest and Rush Hour are good choices

Duck Soup, A Night at the Opera, and A Day at the Races---the Marx Brothers' three best films. (someone's gonna mention Horse Feathers and Animal Crackers, and I'm not going to dispute their valid points, but those are my three. Hell, watch those too.)

Monty Python...and the Holy Grail, Life of Brian, And Now For Something Completely Different, and Meaning of Life.

Slap Shot and Major League for funny sports movies.

Or just put "Yakety Sax" into YouTube's search bar and let hilarity ensue.

TV:
The IT crowd, all 4 seasons. One of the funniest things on television.
Psych. The show has some of the most entertaining and witty dialogue I have yet to see in a comedy.

Movies:
Airplane. The one. The only. The classic. Just watch it. It is brilliant.

Anime:
Ouran High Host Club. One of the most brilliantly hilarious parodies I have ever seen.

Web:
Homestarrunner.com. Very random, but also very funny. Highly recommended.
Red Vs. Blue. One of the longest running Machinima shows around. But also the funniest.

Those are all the ones that immediately spring to mind.

Get Smart, the original series from the 60's, not the movie.

Flying High (Airplane!), probably the funniest movie I've ever seen.

Nothing with seth rogen, so dont even look.
If it wasn't for Seth Rogen and his obvious hand in the movie's plot, Observe and Report would have been really good.

OT: Monty Python as many have stated before me, but if you really want comedy, go look at the thread about stand-up comedy if you like that sort of thing, a bunch of people listed some really excellent performers

Community! Best show on TV, very funny.
Very much agreed! The writing is brilliant and the characters compliment each other very well. You can watch most of the current season's episodes on Hulu, though I strongly recommend starting from the very beginning in order to get a good feel for all of the characters.
Another favorite comedy of mine is the FX Western animated series, Archer. I'm thrilled that it got picked up for a second season, which begins later this month. It's a heavily character-driven and very adult spy/office comedy. Every character is completely over-the-top but with relatable flaws and their dialogue is priceless. I can't count how many times my AIM/MSN statuses have changed because of this show. :3 It's created by the same guys who made Sealab 2021 and Frisky Dingo and is arguably the team's best work to date.
