Two options for me, one that'd never be done and one that really should be.
Kurt Rosenwinkel's Segment from the album Intuit, such a freaking good tune and would probably rival most of the Guitar Hero "hard" songs (hot for teacher, etc)
The realistic one would be Stevie Ray Vaughan's live cut of "Life Without You" off of the album In Step. His solo's like 8 minutes long, and is drenched with soul and ridiculous playing.

Don't Forget Me- Red Hot Chili Peppers (not on Rock Band because of drug content)

And for an explaination because I'm sure I'll be pricked again, Led Zepplin because it's awesome, Tom Waits because I want to put my raspy voice to work, Flynn because of the bass, Fleet Foxes because their the fucking Fleet Foxes. I'd like to see Rio by Duran Duran in one of the guitar hero games. Not that I actually want to play it, but if the guitar part in the game was replaced by the keyboard section in the actual song (as guitar hero is known for doing) I'd like to see some of my friends who are ridiculously good at guitar hero get frustrated as they play the equivalent of the intro to Through the Fire and Flames...during the whole song.
