Yup! I played Okami by not upgrading any weapons, only using the base weapon, and only using brush strokes when necessary, throughout the entire game. No Stray Beads, either. Made the game a little bit more difficult...but not as much as, say...Twilight Princess.

That was still probably the only fault I could find for Okami, was it was too easy.

Doing a no merchant run on Resident Evil 4 is one example I can think of, which wasn't even that difficult.
I did repeatedly used to play the old Tomb Raider games using only my pistols unless the situation truly called for it. I always had far too much ammo by the end of the game.

I actually tried again a few times, kinda a Leeroy Jenkins speed run attempt.
I was doing well on 2 occasions until
1-I got clusterfucked by zombies
2-A Hunter decided to tear my nipples off

Not exactly made harder, but (started MW2 and will continue with others) playing through campaign using only a pistol. Very fun and relatively easy on MW2, not sure how so on BFBC 1 and 2, or AVP though : /

Also, on WoW sometimes I'd just try and solo group quest bosses. Things like lvl 75 elites designed for 3 man takedown, on my own with a pet. Veeeery fun and challenging as a Hunter, as Rogue, FUUUUUU- I mean, difficult, long and tiring but possible :p

I think the only time I did this was when I decided to play Mortal Kombat 2 through with Shang Tsung with the restriction that I would continually shift forms - as soon as a round would start, I would morph to a character, and as soon as the morph wore off, I would morph to a different character, never repeating the same character twice in the same round.

In the first L4D me and a buddy had an idea. We decided to change the game to Left 2 Die by killing Francis and Louis. We actually got all the way to the finale but the second tank got us :(

NSG (No Sphere Grid) challenge on Final Fantasy X - however I lost interest after less than 10 hours, it just wasn't as fun.

All the time, well, as soon as a game gets too easy, I will up the ante.
Also, since i am always streaming videos on my computer/watching tv when I play i'm always at kind of a disadvantage.
Examples:
Tf2: Only bat with the scout (with touchpad) admittedly the bat isn't really a "weak" weapon, but it is hard to use well.
Starcraft zerg caqmpaign: Only use zergling/scourges (failed)
LBP: Playing with my noob friends
Diablo 2: Melee sorc without learning enchant (successful)
and many more

Nuzlocke Challenge

-If a Pokemon in your party faints, you must release it.

-Every time you reach a new route, cave, etc., you must catch the very first Pokemon you see, regardless of your current number of party members. The Pokemon may sit within the box for an indeterminate amount of time as a backup.

-If you fail to catch the first Pokemon you see, either via it fleeing, knocking it out, or having no Pokeballs, you may not catch another. Tough shit.

-This means you can not catch a pokemon in a route you were in before you could catch pokemon. So no route 1, you were there before you had pokeballs.

-If a town you go to contains a body of water or a gift Pokemon like Bill?s Eevee in G/S/C/HG/SS, that may be considered a Pokemon of a new area.

-You must nickname your pokemon so there is some sort of attachment. Each pokemon is a best friend to you, to see them die is heartbreaking.

-If your entire party faints, they all must be thrown away. If you have a pokemon left in a box somewhere, you may continue, otherwise you must start your game over.

-Treat pokeballs as rare artifacts; at no point can you buy more than one from each store. You should only use ones you find in the wild or are awarded.

-You are, under no circumstance, allowed to get a second pokemon from any given area.

-You are, under no circumstance, allowed to use items outside of battle. If you were poisoned on your last turn of the battle, you better hope to god there's a pokemon center nearby.

-You may not switch out before the enemy trainer sends in a new Pokemon.

-You are not allowed to use legendaries in battle.

-If your pokemon is 4 levels higher then the pokemon you are fighting then you are not allowed to attack with that pokemon. (Wild pokemon only. Legendary pokemon acts like a trainer fight.)
