Here's an idea: GLaDOS mentions in the co-op campaign that you're actually "competing with other sets of bots". That would be pretty interesting, actually: A whole slew of randomly-generated puzzles (randomly generated as in a large library of different pre-made puzzle elements that the game decides fits well together) with multiple people (and maybe even teams) attempting to reach a single center point the quickest and then trying their hardest to get back to their area while the other players try to kill them by placing portals at inopportune moments or moving around environmental pieces like walls to smack them, or something of that sort. It would be difficult as hell to pull off, true, but it would be a modern-day masterpiece of a game if they managed it.
I actually like this idea the best of any I have seen in this thread. Maybe even take out the killing part entirely and just see who is the quickest, smartest team/person.

Uh, the guys at valve playtested competitive portal games, and mostly concluded it just wasn't fun.

Apparently, a big issue they had was people getting incredibly disoriented when they fall into a portal, which is bad enough when you yourself put it there, but seriously confusing when it's unexpected.

And, it just seemed not to be much fun for those trying to use a portal as an offensive weapon either.

I don't know. I guess it might work.
But I'd be really surprised if valve hadn't tried it at all.
