Well I see that the Co-optional podcast and Cox'n Crendor has been already covered.
I think you should check out also Let's Drown Out by our very own Yahtzee Croshaw and his friend Gabriel Morton. They recently seemed to have moved away to Let's Plays again but I feel like it's worth giving a shot the thing that already is there. It's more of a relaxed conversation while playing (and later watching) some old game. It might not be some kind of ground-breaking industry-changing podcast but I simply find the dynamic between Yahtzee and Gabe to be very enjoyable.
I don't know if you're interested in non-gaming podcasts but I also really recommend Hello Internet by CGP Grey and Brady. If you don't know them, then CGP Grey has a very popular educational channel. It's kind of hard to really pinpoint the focus of the channel: his two latest videos have been a short primer for the Lord of the Rings mythology and an explanation of the Single Transferable Vote. Now that latter one might sound particularly thrilling, but Grey manages to make it quite interesting.
Brady on the other hand is a journalist, who has a whole host of science channels whose focus ranged from maths to astronomy and physics and more. His videos are generally him conducting an interview with an expert regarding a certain topics (for example the possibility of multiverses).
Now this might make it sound like Hello Internet is some kind of science podecast for brainiacs but actually it' a fairly laid-back discussion of topics from everyday life and they often recommend interesting shows. FOr example it's thanks to them that I found out about the series Black Mirror and also my next recommendation which is the Serial podcast. Serial has so far made it through season 1. It's quite unusual to be honest. It's about a journalist's efforts to investigate a murder case from 15 years ago and whether the convicted, Adnan Syed, was the actual killer. Now, we're talking about an ACTUAL murder case here. Adnan Syed is a real person, currently serving his life sentence in prison and he has become something of an Internet sensation with people on Reddit investigating thoroughly his case.
And the last one is perhaps a weird one, but I just listened to the first of episode of the Deferred Gnosis Podcast. It's between a satanist and an atheist who have discussed the existence of spirituality and whether religion has a place in the modern and future world. I don't know if the satanist part might put you off but I found it personally quite interesting.
Well, sorry for the wall of text!
TL:DR: Let's Drown Out, Hello Internet, Serial Podcast, Deferred Gnosis Podcast (only one episode so far though).

Sleepycabin/Sleepycast is my current favorite, I have been following those guys work for years and I find their silly arguments pretty funny.

If you have never heard them, Jim Sterling's old Podcast work is some of my favorite things ever, the old Podtoid episodes contain some of the greatest discussions ever and Dismal Jesters was wonderful while it lasted. I have subscribed to Podquisition but I haven't gotten around to it yet. Fistshark is also really good, although it really makes me nostalgic for the old Jim and Holmes dynamic.

I've really been enjoying listening to Welcome To Night Vale while playing Euro Truck Simulator 2. It's done up more like an old radio show about a small desert town called Night Vale where paranormal activity is common. There's a long running continuity and you actually kind of get wrapped up in the lives of these individuals. Definitely something worth your time.

Hi People!

My name is Patrick and I'm a player with RPGentlemen, a new roleplaying game podcast. In fact we're so new that we're personally posting in forum threads like this to get the word out to people.

(I can prove it! Yahtzee is pithy! Movie Bob is smart [but wrong about Kevin Smith in my opinion]! I am neither a bot nor a copypasta. I'm sure this is all very convincing)

We're a bunch of improv comedians who love D&D and don't let rules bog down the banter; with two episodes out so far and two more on the way this Sunday, when we'll posting our very first breakaway series! While dungeons and the dragons found therein have a special place in our hearts, we understand that they aren't for everybody, so we'll also be playing:

- Serenity (The RPG based off my personal favorite show, Firefly)
- Numenera (Apocolyptic future-past, set on Earth a billion years in the future)
- Superheroes (Our homebrew campaign that chucks the rules and brings the weird)
- Fairytales (In the vein of Adventure Time, a family friendly show that adults can enjoy)

We're also accepting suggestions/pitches/etc from everyone to be added to our games. Anything from "F*@k off M8" to "Here's my thesis on Dragonborn breeding rituals", so feel free to shoot us a suggestion or death-threat at the place where all our episodes can be found!

www.rpgentlemen.com

Thanks for reading! Sorry for the long pitch, it's my first one and I've had too much coffee.

You just need two: Laser Time, and Vidjagame Apocalypse, both of which can be found here [http://www.lasertimepodcast.com/], along with oodles of other fun content (including more podcasts).

Laser Time is typically a general nostalgia show, typically about things from the 80s to now, the Simpsons, the Muppets, Japan, and older Disney stuff.

VGA is a video game podcasts that has a weekly top five (usually about odd things such as Top 5 Movie Games According to the 1994 Box Office, or Top 5 Games with Child Endangerment), talk about the newest games, gaming news, and a community segment where they read listener's answers to their question of the week.

Both shows are usually done by a group of people that have both worked as gaming journalists and worked in the gaming industry, and have done podcast work before (Most of them worked at GamesRadar at some point).

To show off some Laser Time, I want to share their Erotic Fanfiction Theater (since it's one of their funniest bits, and one of the few segments I can find on YouTube), but since it's technically pornography, I don't think I can (you can probably guess how something titled 'Integrating with Shepard-Commander' would go though). But here's some VGA!:

YES!!! I love those guys so much! I actually contribute to the site.

I'm also going to raise Cape Crisis as one their casts worth listening to.
*hi fives*

Yeah, that's a really good one too. I mean, how else are ya gonna know Henry's gay? ;]

And somehow, I forgot about this Escapist safe Laser Time bit:

But in order to really get that, you have to hear this:

Screw it, let's throw TalkRadar into the recommendation list as well. Granted, it was foolishly canceled a few years ago (with Laser Time and VGA being spiritual successors), so all the topics will be dated, but it's still hilarious.

I'm currently listening to 3 podcasts, 2 of which have already been mentioned, Hello Internet and SleepyCast. The third one is The Biggest Problem In The Universe by Maddox. I find it funny.

if you would like to listen to news and actually have humor than you should try "Wait Wait Don't tell me"
