It would have to be a sandbox game, probably a city-builder or trading sim so that it wasn't dependent on quests to keep the action moving...hmm...

If I could one play one game for the rest of my life, I would stop gaming and go on with my life, I would probably read more comics or see more movies or maybe go have sex with people or something I don't know.
Probablt TF2. It never gets boring or repeditive for me, and to try to get all of the hats for the characters, I would need the rest of my life to do so.

I'd choose Dawn Of Warark Crusade. It still has a large community and it has more mods than I can count.
EDIT:That game never lags, you have a life-time subscription, you can always play it without any problems and the servers are always full.
If i had to pick one to play for the rest of my life it would probably have to be Wow, it isn't my favorite game but even with a lot of modability, you would run out of things to do eventually, and WoW is being constantly updated, and even if they stopped updating it there's still so many people to socialize with and stuff to do.
Little Big Planet 2 - it's already been modded to do Limbo, there's an indication it can do Shadow Complex, it'll also do racing games, and I'm sure it can be set up for tower defense as well, and there will constantly be new levels. It looks to be the first real system seller for the PS3, as it'd be worth it to buy a PS3 just for that one game.
Either Halo 3, Saints Row 2, or Fallout 3. Halo because I love its multiplayer and believe it or not, I could keep playing its campaign too. Siants Row because its just a lot of fun to go around and mess things up in that game. I could play that for a long time before getting tired. Fallout 3 because I FUCKING LOVE FALLOUT 3.

I could probably bare with WoW for a lifetime, as long as there's a constant stream of expansions for as long as it exists.
It's mostly just mediocre fun but the awesome moments are beyond awesome, and strangely hard to get bored of.

Either Starcraft 1 or 2. I haven't played 2 yet but I assume it's fucking amazing but I'll never forget the greatness of the first. And I could play forever with custom maps and bots.
