Conker as a girl...just no.
Lara Croft as a man...it would get rid of Tomb Raider but not much else...
Dante as a woman... there are only a few ways to ruin Devil May Cry. This being one of them.

As much as I love that game, he was pretty much a wimp. Ah well, no one's perfect.

Honestly though, if Sora was a girl, then those crazy fangirls would do lesbian pairings with Kairi, as opposed to gay pairings with Riku. *shudders*

Also, it's easy to imagine the Spiderman Games being replaced with SpiderWoman instead.

Ripley from Alien Trilogy looked like a man anyway (in game and in cutscenes) so would anyone notice?

Jack Valentine from Resident Evil 3 would probably have worn more practical clothing (not that I'm opposed to the sight of exposed virtual T&A) that would explain how he can carry: an assaault rifle, a shotgun, a magnum, ammo for each respective weapon, a first aid box, a rocket launcher and a bunch of ink ribbons all at once without any apparant sacrifice of freedom of movement.
