Losing my Pokemon Yellow save to a glitch when I was seven. As heartbreaking then as it is now. I cried over my Wartortle, floating somewhere in some cybernetic Lethe, for days. :'(

By play, did you mean you beat it? If so,
The scene with Nanako in the hospital. When she died. That was sooooo sad for me.
I gotta say that the animated series really caught that moment. It was damn good.
Oh, that... Good thing she actually didn't.

The only time I've ever really gotten close to crying was in Risen, when those lizards butcher the dog. Why is it the god damn dog?! They could have killed the two human hunters, but noooo...

EDIT: Oh, I can actually think of a human example here. Again, I came pretty close to tearing up. In the first Witcher game, at the lakeside village. That entire storyline involving the two sisters.. yeah, after that had finished and the lakeside daytime soundtrack kicks in, it just made me think in-depth on the whole situation and just how depressing it was. And..
Knowing that the one sister who committed the murder is damned for the rest of eternity for her action that was fuelled by an emotion she couldn't control, tough stuff man.

In the Dragon Age: Origins after you light the beacon and Teyrn Loghain retreats leaving all the Grey Wardens by themselves and then seeing Duncan die made me cry for a good 3 minutes.

Mass Effect 3: I messed up the situation on Rannoch: I didn't have enough Paragon skill and I lost all of the awesome characters. I had nightmares about it too.
Pokemon Mystery Dungeon: I played this a long time ago and I can't remember the details... Something sad happened to a Grovyle but I honestly haven't a clue what it was.

Jorge dying in Halo Reach, that little speech before he grabs you and then you see that his sacrifce was for nothing, more cruisers coming in. Damn, that bit made me well up a little.

I haven't actually cried because of a game (haven't cried in a while at all actually...) but Mordin in ME3 and the end of Portal 2 were the closest. Portal 2 just overwhelmed me with emotions all at the same time.

I hear you-I basically bawled throughout all of ME3-and at the end-since I play a fem-shep with Kaiden, I about cried myself to sleep that night. I know-so pathetic-but that game is so amazing-it's hard to say goodbye!

I don't think I've actually cried in a game, but a few scenes did have me misty-eyed.

Red Dead Redemption has been mentioned. As has inFamous 2's blue ending.

But one that was strangely effective was Medal of Honor, the recent reboot.

When Rabbit dies at the end, and it's a protracted, long, drawn out death, I must admit I felt quite affected. Especially since the game is based on an actual conflict, more specificaly based on the events of Operation Rock Avalanche in the Afghan War. It felt more real than most video game deaths, somehow.

wow. I thought I was the only one. :)

Super Metroid was the big one. The ending got me every time when I was a kid. Other than that, Defcon really moved me the first time I played it, before I got into the "gamer mindset" about it.
