Restoring the first Guardian Sapling in Okami. The game looks really good most of the time, but watching colour and life be restored to an area is just...it's amazing.

Lego Lord of the Rings: The Disco Phial. It's a mithril item you can find by shooting 6 targets at the top of Minas Tirith.
Not only does it serve as an alternative to having to switch to Frodo to go into dark caves, it jams music. Who needs dark moody music while fighting orcs, when you can have this on P2 and watch 'em all dance.
('Tis rather annoying when you get nearby, because you start dancing, but oh well.)

The mission in inFAMOUS 2 that was just a cut scene of Cole and Zeeke taking a breather and having a beer while watching TV. It was just fun at the time but it really drove home the endings, particularly evil Cole's final mission when Zeeke tries to stop him.

Halo Combat Evolved:

A standard FPS starring faceless Space Marine #103934802. Humanity losing a war against aliens, ancient superweapon appears, activate superweapon against aliens. Win.

Then you have the flood level. Cortana is left in the Control room, so for the first time you are alone in a freaky swamp. The standard rock soundtrack is replaced with ambient eerie music, and you find a downed abandonded human plane with a recording about how the crew were massacred. A little further in there's an alien dropship... also downed, something you've never seen before, and the scattered alien resistance is actually scared, you can hear them cowering and crying before you see them. Something is wrong, very very wrong.

Your motion tracker starts malfunctioning, showing allies where there are clearly none, you enter a mysterious structure in the swamp. the deeper you go inside it the more dead alien bodies you see. There's one human survivor, slumped againsta wall, driven mad he attacks you with a pistol babbling random nonsense. Most people probably left him alone, it would have been kinder to kill him.
You enter a narrow hallway covered from top to bottom in purple alien blood; no bodies or enemies, just gallons of blood. At the very bottom of the facility you come across a room, and then the cutscene plays:

And you suddenly realise you're all alone stuck at the bottom of the facility, standing in exactly the same room.

At this point if you pause the game the mission tracker in the pause screen stating your next objective just says one word: "ESCAPE" in all capitals. That was probably the bit that freaked me out the most, it was like they had invaded the very game itself, like they weren't supposed to be there.

Mass Effect one running up the citadel tower while Sovereign sits overhead waving his arms around, that was an epic climatic level to a great game.

Dark Souls pretty regularly had these moments for me, coming across beautiful vistas or getting in and winning a close battle against a stronger foe is an awesome feeling.

Frog Fractions, when you...

upgrade the dragon power and fly into space.

It was just a cute little time waster until then.
You would not believe my face when I played that. It seriously went from :| to O_O.

For me, it was Dragon Age Origins, the Cutscene at the Battle of Ostagar when the Darkspawn start pouring through the woods and the human soldiers are all like "Oh shit, Son."

Because that pretty much was my reaction, too. "Oh shit, Son. This is gonna be ah-MAZING!"

The plot twist in BioShock made me fangush so hard that it wasn't even funny anymore. I was into that game well enough already, but I popped a few veines due to pure excitement when that happened. I was unable to put the controller down at that point.

The horrible part is that I had nobody to discuss the twist with, so after finishing the game I desperately went recommending the game to every entity I knew was at least a bit into games.

In Company of Heroes there's a particularly frantic defense level midway through the campaign. You basically have to hold a hill from MASSIVE attacks on all sides. It's a bit like every RTS base defence level ever, but the sheer gravity and attention to detail of it, the excellent sound design in particular with hundreds of men yelling, guns firing, explosions...exploding all at once, really make this game come alive.

Okami topped itself with awesomeness with each new scene.

"Hey, I can cut stuff in half, cool. Wow, that there blooming animation is lovely. Hey, nice little town. Oh, nightfall. Wow, Shinshuu Field looks like crap. Better restore it. Oooooh will you look at that. Oh, a forest dungeon, how predict - JESUS FUCK A TAO-MONK-LIGHTSABER-WARRIOR BOSS FIGHT! Cool, another Sakuya sapling. Man this dungeon is creepy. Wow, I topple the spider boss with vines and slash its tender parts to kill it, sweet. Awesomeness, I'm riding a log down rapids. AWESOMENESS, Taka Pass! This is almost like a sandbox! Digging minigames! Another Stray Bead! Crimson Helm! Orochi is HUGE HOW DO I KILL IT!"

...

"THAT WAS ONLY DISC ONE?!"

As much as I'm disappointed with Skyrim, Blackreach kicked ass. A great place just to look around in with some really tough enemy's waiting for to make a wrong turn.

L.A. Noire's homicide and vice desks were pretty sweet (not that traffic and arson were not fun, the middle desks just had a much more interesting atmosphere to them).

Call of Duty 1's assault on Stalingrad a la Enemy at the Gates was loads of epic as well; It was the first time I had played a shooter that didn't give me anything to fight back with, and forced you to desperately run from cover to cover. And after that, the charge into Red Square was also incredible.

The moment in Battlefield 1942 where I walked up to a vehicle -any vehicle- and pressed 'e'....

And could drive that vehicle, no matter what it was.

I'm gonna say:

Assassins Creed 1, when you first perfectly stealth kill one of the big targets
Assassins Creed 2, when you kill your first target and Ezio screams out "THE AUDITORE ARE NOT DEAD! I AM STILL HERE, ME!"
Assassins Creed 3, when you begin playing as Connor

Gears of War, when you first encounter the Kryll

Mass Effect 3, when you leave Earth, and everything after that

PROTOTYPE, the boss battles while simple, felt epic

Skyrim, becoming a werewolf

Spec Ops: The Line, basically from start to finish

That's all I can think of right now, but yeah, those instances just made the games awesome

The first fight with the alien warship in Serious Sam 3.

Good god, that fight was glorious. Also, if it's allowed, I would like to put the entirety of Serious Sam 3 on this list. Every single time you think the game can't top itself, it does. The game is the perfect example of how to create the never-ending crescendo that so many shooters strive for.
