The World Ends with You. One of the best RPGs, and thus best games, I have ever played. It has a complex but intuitive battle system (hard to get used to, but brilliant once you do), a great J-pop infused soundtrack, an amazing story with unforgettable characters... all the stuff a great RPG should have.
Elite Beat Agents. A quirky rhythm game with some fun songs, interesting style and really fun gameplay. This one is little more than a time-waster, but it's one of the best ones I've ever come across. I could play this game forever.
The Professor Layton series. Puzzle games with a mystery story woven in, these games present some fantastic, mind-bending puzzles within the context of an enjoyable mystery tale. Great characters and fun puzzles make them fantastic.
The Castlevania series. These games have always been great in 2D, and the DS games are no exception. My personal favorite of the entire series is Dawn of Sorrow, the direct sequel to the GBA's Aria of Sorrow, but the other available DS games in the series are fantastic as well: some of the best 2D action games I have ever played (up there with Metroid Fusion)
The Phoenix Wright series. The recent Ace Attorney games haven't been quite as good, but the first three Phoenix Wright games offered intense and complicated stories with truly lovable and unique characters, all wrapped in a well-designed point-and-click adventure game with some intense courtroom sequences that truly demand logical thought.
Hotel Dusk: Room 215. One of those games for those of us who appreciate the artistic side of video games. The point-and-click adventure aspect of the game is not perfect, but the game is a fantastic little film-noir-style detective story that plays out like an interactive mystery novel. The art style and the fantastic writing contribute to this being a truly unique and unforgettable experience.
Those are a few. The Final Fantasy and Chrono Trigger remakes are worth mentioning as well, but since you said no remakes...
