If I say no, does that mean that I know that I am not sure about anything or does it mean that I don't know if I am sure about anything.

First one is paradoxical and would force me to say yes. If only because I would be sure that I did not agree with option b.

"I think therefore I am" is the only thing I am 100% sure of. The nature of my existence could mean that literally everything is an illusion, or the world is as I see it. Everything else is up for debate.

There is something you can be sure of using epistemology, though. The existence of your own thought process is proven by considering it. This is, basically, the only thing you can be sure of being absolutely true.

Any prediction cannot be certain with 100% accuracy.

Edit: A large number of people are posting "That God doesn't exist", which of course cannot be understood as a 100% certainty, mainly because things like God are non-falsifiable. It cannot be demonstrated that it does exist, and it cannot be demonstrated that it doesn't, thus leaving a gap in certainty.
