Uncharted 2. MGS4 was good, a great game, but it wasn't fun to PLAY. It was cinematic, it was necessary and it was a great end to the Metal Gear Solid series, but even with the "improved" controls, it was still clunky and I'll be honest and state that I enjoyed the cut-scenes more than the parts you played.
But Uncharted 2 - cinematic AND a joy to play. Yes the story isn't deep, but it's well written with a cast of likeable characters. It was my favourite game of 09 by far.
However, Uncharted 1 wasn't so great. It wasn't bad, but it shouldn't be on this list.

Little Big Planet. I don't have a PS3 myself, but I play it at my friends house all the time. I love customization, and Little Big Planet murders the forge mode in Halo 3. I'm really getting in to editing Amnesia maps, but I can't get them to show up in the game. I've already designed one with a cfg and lang file (nothing else) and I can't get it to show up. It's in the file, but does it have something to do with scripting?
Uncharted is becoming the face of the system, Uncharted 2 is pretty much flawless. MGS and LBP try their best, but right now Uncharted series is hard to beat.. also Gran Turismo will hopefully get on lists like this whenever it decides to come out.

This question, which I do not like, reminded me of something else, which I do not like. The PS3 is full of action and shooters. Where's the strategy/rpg love? 
Truthfully, PS3 is best 'represented' by an action or a shooter so I can't answer with Demon's Soul(which is a hybrid action rpg?), which I liked.
