BestBuy for new games, GameStop or Half-Price Books (really really really cheap, all the games I got there were under $10) if they have a really good old game.

Gamestop mostly on rare occasion I will buy from Movie Trading Company or assorted other places like that.

It depends what I'm looking for and who happens to have the best price at the time. Lately I've been buying most my games on Steam, or this onlineshop for my console-needs.
I also tend to pick up games I've wanted a while when I spot them in the bargain-bin so to speak.
I'll only buy used when there's something I can't get a hold of any other way tbh.

Steam or Play.com, depending on the platform.

Steam, obviously, for my PC games and I get most of my PS3 games from Play. I'll occasionally buy from Game or HMV though. It depends who have the better prices.

Almost exclusively GAME for consoles. I've got a fuckton or reward points from my spending spree that stretched over the last two months. I have been known to buy from Discs & Disks on rare occasions; if they have any awesome special editions leftover in the stock room.

Bioshock 2 Special Edition - £25 get in!

Anything for PC usually gets bought over Steam.

I buy from Gamestop with the added modifier that one of my local gamestop stores is managed by a close personal friend of mine. So, even though I'm allegedly betraying the whole industry, I'm making sure someone I know keeps his family fed.

D2D is by far a cheaper solution than Steam though. I'm talking above and beyond 40% cheaper.

Steam is only good for sales, and even then they're not always cheaper.

I buy online most of the time. Amazon, CDON and Play-Asia (and eBay for out-of-print stuff).

EDIT: + Steam, naturally.

Gamestop for new releases/preorders. Steam for "non-anticipated" games, generally with sales. Amazon for hard to find games.

Gamestop cuz its close by and has older games (like everyone doesn't know that already).
Steam if I want something for my PC.

Play.com for games I really want with new releases soon (Like White recently) Amazon, if I can't find the game (generally for stuff like the yugioh world championship games) GAME if I'm in town, and recently, a new game shop that opened in town called totally games.

This poll is skewed towards the americans I've noticed, I don't think there is a gamestop in the UK, correct me if I'm wrong.
