Star Trek and 40k have both already had a lot of shots, some good, most bad, but they've had chances. Since the world needs an honest to god tactical fleet game, or a really interesting space fighter game, or a bioware-made mass effect-styled adventure game would all be fun. Add to that the fact that there's just a lot of potential things to do with a B5 game. Set it after the serieses, and it becomes a 'find a cure for earth' game, set it before the series and there's the centauri/narn war, the minbari war, any of those. There were enough episodes with events that you could reasonably make a game that just took place during the actual series itself. The second centauri/narn war, the shadow war, the minbari civil war, etc etc. A real fleet game set like that- aligning your ships along the best attack axis for your weapons complement, arranging battle plans, redistributing power systems, etc would be really fun, even if it only centered around a single player one-faction campaign.
Or if it were a god's honest space fighter sim, where constant thrust and velocity weren't synonymous, where it wasn't a flashy, splashy frenzy of particle effects and light making enemies impossible to target. Hell unlockables for that like a Shadow or Vorlon fighter would be utterly reasonable and could even allow you to incorporate player-decision-story-affecting bits. Like if you play as a good guy you get a vorlon ship. That's just my opinion though.
