Assuming he can be considered an assassin, I'm going with Ryu Hayabusa.

As for wetboys, well that's an entirely different matter.
If we were talking wetboys, I'd have to say Durzo Blint...

Paraphrasing....

1: "Out of everyone in the whole world, who is the most effective assassin?"

2: "Well, I would imagine it would be one who would be able to conceal their identity as an assassin."

1: "Yes, but I can think of an even more...ideal assassin. One who does not know he is an assassin..."

Name the movie!
Naked Gun. :D

OT: Going with 47.

Damn, ninja'd.

Ironically, Ninjas make the best assassins.

Dis counting the Other category, 47. Because he has more than a crappy pistol like Ezio. He has friggin' silverballers, not to mention a silenced shotgun with thirty rounds in Blood Money.

"Assassins have targets, wetboys have deaders."

*Ahem* I think it's safe to say Kylar before he learns magic.

The Red Spy.

He has a watch that makes him invisible. A Magnum for sir, and a Butterfly Knife for madam.

Best of all, he has a set of masks that make him blend in seemlessly.

47. His only identifying mark is his barcode whereas the others come with their own team jerseys. A huge portion of assassination is stealth.
Yeah, bald with a barcode and pissed off face certainly doesn't alert people, especially when they patrol the same areas so you'd think they would notice when 'bob the janitor' looks completely different and runs around important people suspeciously.

Not that I'm complaining, love the Hitman games.

I'd go for Karasu for stealth (Shinobido), no social stealth so its fairly straight forward.

Edit: Fuck that, I'd go with Garret.

Also that wasn't actually an edit, I just thought about it after I wrote the rest.

What? Never heard of him? Exactly...

Oooh you mean in games.
Tenchu 2's Tatsumaru.
The rest (excluding 47) are absolute pussies.
Eizo/Altair would not even know what was happening until he snapped their necks and urinated on their rapidly cooling corpses.
Considering he's from a tenchu game, meaning he ways roughly a ton and carries himself as such, I find that hard to believe. Not to mention that the enemies will believe that a duck or dog clearly killed their pals, even if they had just seen a ninja run through. (Not that AC lacks similar flaws)

Seriously, that game was so slow (fun when I was 12, but slow).

PS: Loved using that red demon guy who'd suicide bomb in the VS matches.

Ezio because he had a personality and some character development.

Not just "I used to be an insensitive douche. Now I'm a slightly less insensitive douche"
That's his weakness. He acts on emotions. 47 is the best assassin. Nothing personal, always business. And he's pretty good at what he does.

Considering the arse-nal and setting, definitely Ezio. For the guys saying he gets seen killing, that's really the fault of how the game is made. In Hitman, levels are designed in a way that pretty much ALWAYS leaves your target alone at one point or another... save a few exceptions, but in that case you usually just use a sniper rifle, which is kind of a technologically unfair advantage to compare with Ezio.

If AssCreed levels were designed like the Hitman ones, Ezio could pull off never being noticed easily. Also, to quote Leon... the sniper is an amateur assassin's weapon... as you get better, you use shorter and shorter range weapons - the last weapon you learn is a knife (although to be frank, one of my definitely favourite weapons is a sniper rifle, assuming a nice position, there's few things more satisfying than taking a target down without anyone knowing who got him or even remotely where he is)

Also, note that AssCreed post-assassination guard chase is pretty much scripted. There are loads of levels where you can go by completely unnoticed, have no one paying attention to your target, but the second the blabbing deathbed ends, every guard in town has you pegged as an assassin.

Altair, I choose you!! (sry, been playing waaay to much pokemon)

Other than Altiar, It'd have to be Rikimaru, from the Tenchu: Stealth Assassin games on PS1 and 2. He was bad-arse, looked the part and was super deadly. What's not to love??

I still maintain that it's the best ninja game around, tho we need a new one badly, because I don't want to have to go to a 12 year old game to be a proper ninja.
I tuned out on ninja's in games since they started wearing safety orange.

Ezio has a gun but doesn't need to use it, enough said?

Really though got to go for Ezio for the amount of ways he can kill his target, he's kind of like the Subway of assasins.

Altair and Ezio aren't meant to be stealthy. Their style of Assassination is to show up from nowhere, kill the enemy spectacularly, then vanish into the crowd. 47 kills people because he's contracted to take a life, they kill people to send a message.

Of course, in the real world their way would just get them killed quick, so I'm gonna say Sam Fisher.
