Its a bit hard to use a blade when you cannot breathe and are being held above the ground.

It's really hard to choke someone when every time you mention his late wife's name he hits the floor in the fetal position =p

If it was a mid-range battle: It could go either way
If it was a close-range battle: Vader (Lightsaber would cut through the masamune like butter and Vader has force powers)

About Seph's speed: That's irrelevant since the force will allow Vader to predict any move, no matter how snappy. And Masamune is no match for a lightsaber. Vader only has to block it once and it's in twain.
