I already have:

Devil Survivor. JRPG from Atlus. Surprisingly deep tactical combat, one of the best nonlinear story structures I've seen in an RPG, epic soundtrack.

Castlevania Order of eclesia (I heard all 3 Castlevania for de ds are good)

Phoenix Wright: Ace Atorney (This is more of a layers/investigation text adventure/point&click game. Its good I promise, just weird. Also very funny)

Elite Beat Agents(This is a must if you like music games, a little short but a fun ride while it last)Also rythm heaven.

Advance Wars: Day of Ruin (a turn based war strategy game, with a post apocalyptic setting)

Mario vs. Donkey Kong 2: March of the Minis(A lemings kinda game with touch controls)

Oh and if you like 2D mario dont forget to get New Super Mario World.

999 was very interesting puzzle game throughout.
Flower, Sun and Rain. If you like Suda's style and humor I recommend, though it does get boring at times.
Etrian Odyssey. Really enjoyed playing it even though I only got to level 25. Very oldschool RPG.

1 Any Professor Layton game althought the first game is super cheap now.
2 The World End With You
3 Hotel Dust (Point and click/ detective type of game so it may not appeal to you.)
4 Infinite Space (It's a Platinum game so you know it's good since they did Vanquish and Bayonetta.)
5 Dragon Quest IX (If you like to customize your characters) or Golden Sun Dark Dawn.

Actually, that's a pretty good start. What else can I recommend?

Mario & Luigi: Bowser's Inside Story. Mario and Luigi go on an RPG-style adventure throughout Bowser's body. Meanwhile, Bowser works to take back his kingdom. The battle system is pretty fun, and the game has a great sense of humor.

Phoenix Wright: Ace Attorney. First-person adventure game where you play as a defense attorney trying to prove the innocence of his clients (and they usually are innocent). There are several games in the series, but I recommend this one because it's the first of the series, even though I like the second and third ones more. Just try it out and see if you like it.

Ghost Trick: Phantom Detective. You are a ghost trying to solve the mystery of why you were just killed. You have the ability to possess and manipulate small objects, and you can also travel a few minutes into the past to save other people from death. It's a pretty fun puzzle game with a funny and interesting cast.

Pokemon HeartGold Version. Personally, I think I kind of like this Pokemon game more than Black or White mainly because of two things: 1.) it comes with a special pedometer that lets you obtain Pokemon when you use it, and 2.) You actually have uses for the touch screen. You might want to try Pokemon Black first to see if you like the series, then give this one a shot. (I would also recommend Platinum Version for this slot, too.)

Mario Kart DS. This is probably my favorite Mario Kart game that I've played. It has a rather wide variety of courses (32) you can race on, as well as having online multiplayer to race with people around the world in addition to local multiplayer. It also allows you to play Battle Mode (which used to be my favorite way to play Mario Kart) even if you're alone, which most other games in the series did not do yet.

"Hotel Dusk: Room 217" and "Last Window: The Secret of Cape West" are amazing DS titles, and you can play either without the other (but both are great!). Recommended if you love strong story-lines and well-made point-and-click adventures. If I had to choose between the two I would say go for Last Window, because they fixed all of the tiny things that bugged me about Hotel Dusk.

"Geometry Wars: Galaxies" is a really fun arcade-style shooter that's perfect for pick-up-and-play. I would also recommend "Pokémon Link!" (or "Pokémon: Trozei!" in some countries) for the same, especially if you're a Pokémon fan.

"Sonic: Classic Collection" is a nice title to have, but I felt kinda dumb paying full price for it, so I'd recommend second-hand or in a sale.

"Mario Kart DS" is the quintessential driving game for the DS. It utilises the controls really well, the graphics are great and there's hours of entertainment there.

I'll also say "Pokémon HeartGold and/or SoulSilver". "Pokémon Black" is actually my favourite Pokémon game in the series, but these come in close second for their lovely enhancement of Gold/Silver/Crystal, as well as the clever use of the Pokéwalker.

Castelvania: there's 3, pick one, all are good, or don't and get all 3

Mega Man Zero Collection: 4 games in one :D all are great

Devil Survivor: well anything 'SMT' will serve you well really

Chrono Trigger: cause, Chrono Trigger

only 4 but eh, alot of the other good ones are already up there

Etrian Odyssey 2, Etrian Odyssey 3. Awesome classic Rogue-like RPG with an emphasis on dungeon-crawling and map making. (EO1 was broken in terms of balance, which was fixed in EO2. And, EO3 uses a whole different class system.)

There are a lot of other awesome titles as well as those, DS is a great system. Or rather it's an awful system with a great library.
