Definitely Cyanide and Happiness. Those guys are hilarious.
Also, Axe Cop is pretty amusing too if the humour is in your tastes. I love it, so...

Questionable Content without a doubt is one of the best i've read. but if your new to it, it takes fucking ages to catch up, its almost hit 3000, strips right now and has been going for about 7 years now.

If you're into story-based webcomics, then 'Gunnerkrigg Court' is awesome and definetely worth a look.

It's being going for a while now, and the author updates three times a week so there's a lot to read though. But the over-arching story is great and full of plot-twists and mysteries, the characters are believeable and everything progress at a really good pass that the author reallys shows in the art style.

Plus it has a good grounding in British mythology, among others, so I already love it. :D

Also good are 'Exiern' and 'Yellow Peril', which are both great story-comics.

And for the love of God, 'GRIM TALES FROM DOWN BELOW'! And Snafu comics in general...

xkcd is fantastic and well worth checking out although most folks have already mentioned that.

A Softer World is also well worth a look in although the humour tends to be a little more heavy handed.

If you're a Nintendo fan theres nothing better than Brawl In The Family, and also Awkward Zombie but that has a few comics to do with WOW aswell. If you're not then I dunno, I don't really read any others that you haven't mentioned.

And my current favourite... [link]The Guilded Age[/link]. The story has gone in an odd direction lately, but I'm so invested in the characters and the new spin is so interesting that I check this one religiously. And they update regularly, YAY!

Others I recommend but do not read as often have already been mention here several times over. Oh and even if Hyperbole and a Half is a humour blog and not a webcomic, I join the ranks of those heartily recommending it.

Read The God of Cake or Dogs Don't Understand... without laughing. I dare ya. And this is me down to a tee.
