Wayward Sons is cool. Freak Angels too. They both are serious (with occasional bits of humor)and have great artwork.

Great characterization, high quality art, continuing story arc, and The Cock of Science [http://templaraz.com/?p=302]. Just, if you get to the end, try not to worry too much about the spotty updates.

On the lighter side:
Darwin Carmichael is Going to Hell [http://dcisgoingtohell.com/]
A stylish, brightly colored and light-hearted world of fantasy creatures and humans just getting on with their lives. It starts a little slow, and it's not always hilarious, but it just feels good inside. And it's the one bit of cuteness I can deal with that isn't blown out of proportion like most internet things.

Some of my favorite completely-imagined fantasy stuff. Again, slow updates and slow-moving plots, but once the story's done, read the hell out of it. There's a good deal of finished content right now.

And if you don't mind the fact that it was origionally intended to be porn and as such has a lot of nudity, sex and crass humor, Oglaf, it doesn't really have one long strory, rather a number of smaller ones with a few unrelated one shots, it's also mostly a gag comic.

Lackadaisy. Stunning artwork, fantastic range of characters, great development, charming, funny, witty, stylistic and nostalgic, plus it's an ongoing story, not just a gag comic.

The Abominable Charles Christopher. Great ongoing story along with, for my money, artwork as original and clean as you could ask for.

If you like fantasy, Errant Story. Great art, great story (one of the most story driven webcomics I have ever read)

Again, if you like fantasy, Flipside. My avatar is the main character. Also great story.

And, yet more fantasy, if you like drow then Drowtales is an excellent webcomic. It actually makes the drow make sense. It is richly detailed and the plot is quite good, if rather complex.

Finally, if you like D&D specifically, then Order of the Stick is excellent.

... yeah, I like my D&D inspired fantasy webcomics, don't I?

If you don't mind something different, try Platinum Grit [http://www.platinumgrit.com/]. It is a fantasy/sci-fi comedy based around a masochistic nerd who chaperones a vindictive chick. They inherit a castle in the Scottish highlands, and get up to wacky hi-jinks involving aliens, magic, pagans, feminist journalists etc. Very funny and extremely well drawn.

If you've already tried Questionable Content, you could give Something Positive a try. Be warned that Milholland can get pretty off the deep end sometimes. Good story and recurring characters.

The Meek is brilliant, but it doesn't update often. It fits all your other criterion.
Other than that, Ratfist and Dr. McNinja. They're both humor comics, but they are very well written/drawn and tell an ongoing story.

A couple people have reccomended it already but I'll say Freefall. Another good one is Paradigm Shift. Not a humor comic but one of the best suspense stories iv'e ever read.
