Preferentially, you may only ever want to play it once or twice.

Depends on the game, and on the DLC. Which DLC for which game are you getting? Say, if you completed Shivering Isles in Oblivion, you could only repeat the quests if you started a new save file, or if you downloaded the DaVinci quests in AC: Brotherhood, you can replay them from the pause menu.
