All the cheesy jokes in eat lead, yeah i played that stupid stupid game and loved it,
Also going to have to say my blind semtex out a window, trying for the close-enough kill with danger close and i stuck the poor bastard, ohh what i would have given to see his kill-cam replay. ^o^

I remember when my roommate was playing through heavenly sword for the first time, and the main evil dude did this funky, batshit insane laugh and I about died. Or anytime that psychotic twing twang girl said anything was also pretty hilarious. Other than that, most my laughs come from griefing kids in multiplayer like the time a chased down a kid with a riot shield in MW2 over half the map cause he couldn't figure out how to kill me.

GTAIII and San Andreas definitely had some awesome radio. Maybe it's just me, I didn't really like GTAIV's radio stations.

When you first get to the part with the giant pistons. "Didn't we have some fun, though? Remember when the platform was sliding into the firepit and I said 'Goodbye,' and you were like 'NO WAY' and then I was all 'We pretended we were going to murder you.' That was great." This was, without a doubt, the best line in gaming history.
All the Recent Sam and Max seasons. Those two and all the crazy characters keept me chuckling the whole time.
The easter eggs is almost anything give me a good laugh. Most recently it's been the Metal Gear Solid easter eggs, specifically 4.

A few things, some of which I probably shouldn't admit to.
The 'probing Uranus' easter egg in ME2, the 'witty' banter between Cole and Baird in GOW, and not so much in a game, but during an online match, the temper tantrum my former lecturer threw when he was unceremoniously killed and proceeded to rant about gaming conspiracies, lag time, BT internet, and us being a bunch of f#cking c@nts because we were all 'cheating' and conspiring against him. And I was his teammate.

Video games can rarely make me laugh, but when it does it's pretty damn awesome. I've had several moments of lol's irl, from Borderlands, Zombie Island of Dr Ned(Scooby Doo Parody, and Dr Ned himself) and from ME2(the implications of beastiality happening on the ship from Mordin. )
Deb of Night, the radio program playing in Vampire: The Masquerade - Bloodlines. Hilarious, particularly the commercials.
KotOR II. HK-47 in particular had me reeling from laughter.
Dragon's Age: Origins had me laughing a lot, too. The banter between teammates is usually hysterically funny, and my dialogues with Shale was great, too.

ODST, in a solo Firefight on Lost Platoon, i was dodging a Brute Chopper and stuck it with a plasma grenade and he jumped off before it detonated, he survived but the bike exploded and one of the wheels smashed right into him for a hilarious surprise Splatter medal.
Thinking I have a throwing knife, tossing it and stuck a dude with a Sentex on MW2. Hearing a Salarian scream on Mass Effect 2, using the Singularity heavy weapon. Sent flying halfway across the map in Borderlands when an enemy crashed his car into mine, causing me to get tossed out in the process(no health was lost, though the fall would have killed me).
