Good old fashioned surprise-emotional manipulation n______n. The standout one is of course in Fullmetal Alchemist with Nina.
I dunno, FMA started out with a hideously failed experiment, 2 deaths and an undeath. I don't think you can really say that show isn't supposed to be serious, even if it had its lighter moments.
[sub][sub][sub]Bloody thread title [/sub][/sub][/sub]

I guess you got me there. I was thinking more of a whammy moment but that's laziness on my part, I apologize.

So many examples people have already said, Will's confession in the "very special episode" of Fresh Prince, the episodes "jurassic bark" and "Luck of the Fryish of futurama"...

If I had to pick the single most surprisingly emotional response I had to a show that is anything but serious, though, it would probably be adventure time.

I don't really follow the canon of the series, I just watch the episodes if they happen to be on because they don't take much thought, and are worth a few cheap chuckles.

Then along came an episode where Ice King asks Marceline the Vampire Queen to help him write a song so he can impress dem princesses, which is a fairly standard episode plot, until it leads into a completely unexpected reveal about the past the two characters share together, complete with a duet that leads into a flashback that ends the episode.

I admit, that song made me tear up, and I didn't even think I had any emotional investment in the show.

The Holmes and Watson sketch, the very last sketch in the show, from That Mitchell and Webb Look. Following four series of hilarious sketches, it really caught me off guard. Watson visits a senile Holmes, pretending that Holmes is as sharp as he ever was. There's a few laughs here and there, but when Holmes says 'I know John, I do know. I can't get the fog to clear', I just wasn't ready for it. When the credits rolled I just sat there devastated.

I'm gonna say the final scene of Blackadder goes Forth. Hell, I'd even go so far as to say the whole final episode.

George's reluctant: "I don't want to die..."
Blackadder's: "It's got to be better than my plan of pretending to be mad. Who'd notice another madman around here anyway?"
The final scene of the platoon going over the top with the melancholy rendition of the theme tune fading into a shot of a poppy field. That's made all the more remarkable considering that the ending was a bit of a fluke, their original ending was terrible.

I'll take the easy answer;

Jurassic Bark; Futurama (Season 4; episode 7; the episode with Seymour the dog)

If you ever question if your friend is actually a demon, show him/her this episode and see if they cry at the end. If they do not, they do not have a soul and therefore are a demon.
I have no idea why people always mention this episode of Futurama. Sure, it's touching, but I think "Luck of the Fryrish" is much sadder. "Don't you... Forget about me..."

Specifically the scene were...

...Eric is about to kill his own mother, and is then shocked by the realization of what he's about to do.

This song about a cartoon platypus missing his evil scientist nemesis made me cry:

Phineas and Ferb has many such moments, actually, most of them having to do with Dr. Doofenshmirtz (although maybe I'm biased.) and his various issues.

Also his giant robot Norm. Especially 'Real boy', where we see how

He looks up to Doof and sees him as his father. While Doof treats Norm like junk, not even realizing how he is treating Norm the same way his parents treated him, and letting the cycle of neglect and abuse continue.

EDIT:

I think it was inspired by this:

I'm pretty sure they mentioned the ending to Blackadder in the series and there was a sketch on how they'd end the series with a tragic moment.
Also the sketch-series on the post-apocalyptic game-show in 'That Mitchell and Webb look' was both funny and horrifying, and by the end it had a bigger impact on me than the Holmes sketch did.

Futurama has, at least, an episode with a touching ending per season. Other examples include "The Series has Landed", "The Luck of the Fryrish" and "Godfellas", but "Jurassic Bark" is still the best.

OT: The Fresh Prince of Bel-Air has several touching moments...

The pilot episode of Community, where Jeff was sitting on the steps of the library(?) and his line about 'The funny thing about being smart...'

That episode was funny, but that scene got me hooked on the show

AND NOW HE BROKE MY HAND!

Well it's easy to see why people would flock towards Jurassic Bark over Luck (though both have had shout-outs so far)

Everyone loves dogs and therefore it's sadder :p

Mild Snakes on a Plane spoiler for the two of you who haven't seen it but are really excited to see it this weekend:

Snake eats the dog: Oh my God! That poor dog!!! What a horrible thing to happen!
Snake bites a mans ding dong: BWA HA HA HA HA HAAAAA!!! He died! :D (or the dozens of other human deaths in the movie)

Various S Club specials.

"Boyfriends and birthdays" comes to mind. Hard to put my finger on why...I think it was because they keep making the same sort of rubbish jokes they always make, only the upset people are acknowledging that they aren't funny.

And there's a kid who is all excited that it's his birthday, and that he'll get to see his father that he hasn't seen for years. And people are trying to quietly drop hints that the reason he's not seen him in years is because the father doesn't care, but the kid just refuses to recognise it. Also, the dad doesn't turn up in the end or anything like that, no cop-out happy ending.

...

Also, the ending of H2O Just Add Water.

Bella ends up with her love interest, Cleo ends up with hers (who came out from the US to be with her), Rikki doesn't end up with hers cause he's a tosser and she's not taking him back. Normally, I'd expect to see some crap "happy" ending, where she forgives him and all is nice and all...only that's rubbish, and I'm sick of it.

Yeah, bot kid's shows, but Blackadder and Jurassic Bark were taken.

Easy ones have already been mentioned.

And though scrubs has been mentioned I'd like to point out a specific one that really got to me. When Jordan's brother returns, turns out to have something incurable, and dies. And then what Dr. Cox goes through, and the ending. "...Where do you think we are?"

And I'd also like to point out that a later episode in Futurama kind of gives a semi-happy ending to Jurassic Bark, when we find out that the Fry that got sent back to the past, had his voice changed by an accident, and lost his hair, actually returned after those 2 years of Seymour waiting for him. Which fits more with the title. As in "touching" instead of "sad" moments. :p

Watmote got me a couple times in the later episodes. I just felt so bad for the girl being THAT bad at human interaction. The pity led to a couple pretty sad moments.
Y'know I felt the same way but after thinking about it more she really is a shitty person. She's so arrogant and the things she says about other people she knows nothing about... but I suppose crippling anxiety can do weird things for you.

Speaking of Community, Abed's documentary episode really hit home with me, being autistic myself. Didn't expect an episode like that to show up either so it caught me off-guard. Made me choke up too, which is a big deal considering I'm pretty damn stoic.

Not forgetting the scene before that, where General Melchett tells Captain Darling that he's arranged for him to be transferred to the front lines, so he can take part in the attack and get his share of the glory. True, it's played for laughs, but we're still watching a man on the verge of tears as he's sent to certain death by a senile old bugger who genuinely believes he's doing his friend a favour.

Add "Leela's Homeworld" to the list, especially the flashbacks showing Leela's parents watching over her, and the lengths they were willing to go to spare her the stigma of being descended from mutants.

In Sayonara Zetsubo Sensei, one of the main characters, Komori Kirie starts to stalk her teacher... like all the rest of her classmates, even the male ones. However, while most of them (except from one, whose name means "to follow around; to stalk", both of whom follow him into the toilet) she eventually tells her teacher that she's been taking care of him and his nephew, and that she's grateful for basically giving her a reason to come out of her lair (she's a huge agoraphobe) and finally make friends.

It's not particuarly emotional, but this is also a show where 90% of the dialouge can be summarised as:

Teacher: I've lost all faith in this society!
OCD girl: Then do it some other way!
Creepilly optimistic girl: Come on everyone, be happy!
*everyone is happy*
*Big-boobed Russian girl flashes panties*
*credits*

Also the end of the manga/show, which was also quite emotional. I'm not going to spoil it, though, even though it's been foreshadowed very heavily from the very start.
