Of all time? Really hard to pick one. It depends what I'm looking for in a show. I'll give you a few of my fave picks though:

The Big Bang Theory, Bottom, Scrubs and Arrested Development for comedy covering most tastes (Geeky, slapstick, mainstream and um, different, in that order)
Doctor Who / Firefly for my sci fi fix
Walking Dead, horror, obviously
House and Veronica Mars for my drama (both of them still make me laugh/smile too)

Hopefully that's enough to go on for now!

Edit: Oh, and Russell Howard's Good News and Rhod Gilbert's work experience have both been excellent.

The Wire, The Sopranos, Mad Men, Boardwalk Empire, 30 Rock, South Park, Scrubs, The Inbetweeners and there's plenty others out there, so it all just comes down to what your tastes are.

It's not my favourite either, but I'll back this up.

My two current favourites are House and The Chicago Code.

The Avengers (especially Series 4 & 5 with Diana Rigg)

There are other shows:
Burns and Allen Show (classic 1950s comedy with a great comedy team)
The Wire
Rome
Veronica Mars
Battlestar Galactica
China Beach
I'll Fly Away
(and more)

But...If I have to pick one show only? It is The Avengers.

The Twilight Zone. I love lots of shows, and my first reaction was to write The X-Files, but even that show borrows a lot from the undeniable king of sci-fi, suspense, and horror. It's also aged remarkably well.

Futurama, because everything about it is fucking awesome. Humor that gives even golden-age Simpsons a run for its money, extremely developed characters, handling drama with finesse, and airtight continuity. For someone who has always favored animation over live action, the first four seasons are a dream come true. It's one of the very things things out there that I could call "perfect".

Scrubs is one of the best shows I've ever seen. > Owns seasons 1-8 DVD's

However, like mentioned previously, Avatar The Last Airbender and MLP are pretty fucking awesome.
