Hmmm... As far as I can remember, it was the destruction of a Combot in Metal Fatigue..

Though I could be wrong..

EDIT: Actually, I think I might be wrong on that notion..

I think the first enemy I killed was a Zergling.... I was born and raised with StarCraft!

A wolf from a turn-based LOTR Fellowship Of The Ring Gameboy Advance game. I killed it with sticks by sending my characters back and forth, running penguin style, to batter in the poor mutt's skull. It was glorious!
