I have to go with Skynyrd. But I'm having trouble even thinking of serious contenders against them.

Not to take anything away from Tool, but greatest ever implies a certain amount of longevity.
Tool has already been around for 20 years for your information. Although Tool is definitely my favorite American band, I would have to give the title of greatest to Aerosmith.

Does it? Well, music is subjective anyway =P All a matter of opinion! I thought greatest was just completely awesomest... anyway I change my vote, I vote Black Label Society now!

20 years isn't long enough, and besides, not everyone loves them, or remembers them fondly. They just don't say american to me.
I think Grand Funk Railroad is the Greatest American Band. Tom Petty & the Heartbreakers. Now that smells of apple pie.

Well, let's just make a list of potential candidates:

Lynyrd Skynyrd
The Eagles
Jimi Hendrix (my vote, btw)
Metallica
Aerosmith
Poison
Guns N Roses
Boston
Pearl Jam
Nirvana
Van Halen
ELVIS PRESLEY (seriously, can't get much greater than that)

There is, of course, a lot more. This was just from a cursory scroll through my iTunes library.

...does Rush count? They're Canadian, which you could say is 'american'.

What were you saying about no serious contender to Lynyrd Skynyrd?

EDIT: right, the post above me reminded me of some more: Bob Dylan, The Beach Boys, Tom Petty.

To stray away from the rock stuff, though: Michael Jackson, The Temptations, Robert Johnson, Son House, The Supremes, Smokey Robinson and the Miracles, Stevie Ray Vaughan, and much much more.

Grand Funk Railroad. Definitely.

Nothing is more "apple pie" than them (Except for actual apple pie).

I think it might be difficult to pin down a specific artist due to the many genres and tastes within the music industry. That said, I might be inclined to say Metallica, Guns and Roses, or indeed Lynyrd Skynyrd. But did any of these really have the mass likeability needed to be great?

I'm gonna nominate Disturbed for this award.
that is just stupid. By far, the greatest american band is probrably metallica, nobody, and i mean nobody has had a greater impact on the american metal scene in such a short period of time.

Boston, Aerosmith, and maybe Metallica could qualify for nomination. Michael Jackson and Bob Dylan aren't bands, they're artists. Phenomenal, amazing artists, but individuals. What I'm looking for would be like an American answer to the Beatles.
