WARNING: the actual answer to the question, relayed to me from someone allegedly 'in the know', follows. You may not actually want to know this, as it is not in the film for a good reason. The briefcase contains Marsellus' soul. Hence the importance of the package to Marcellus and the white light, etc. This part of the plot was excised from the movie somewhere between the initial script drafts and the final print because it was considered a bit too "ghost-story-ish" and would be received poorly by audiences who wanted a more gritty, realistic film. Also, as previous poster has pointed out, it's not really necessary to know what it is. Not knowing also adds some mystique to the film.
You'll notice that Marcellus always has a band-aid on the back of his neck, this is related (i.e the audience is meant to imagine that perhaps his soul was extracted using some kind of witchcraft or other method that left a lesion on the back of the neck).
Sure, QT has said this isn't true, but then he wouldn't admit to it anyway, because admitting to it defeats the whole purpose of not putting an explanation in, in the first place.
