Worms series anyone? I freakin laughed my ass off at the horrible things that happened to those poor guys, especially when i orchestrated their destruction. I still remember making the worms into a pyramid and detonating a holy hand grenade in the middle, good times.

The only games I can think of that had me laughing out loud (non maniacally) are Portal (1 & 2) and Fallout New Vegas, in the Old World Blues DLC.

Oh god yes Old World Blues... That was just fucking hilarious. Love that DLC.

Conker's Bad Fur Day simply because of the Sloprano which just came out of nowhere and was both fantastically wrong and hilarious.

No love for the Monkey Island series? Well ok I've only played Curse of Monkey Island but that game kept me giggling or chuckling non stop throughout the game.
Sure I've played some games that had funny moment but that game funny moment was all of it!

Not only does it supply it's own humor, with an ungodly amount of references to pop-culture and down-right ridiculous scenes, but killing each other is funny as hell.
Also, Fallout 3/NV have a dark scene of humor I really love.

The World Ends With You, without a doubt - it's even funnier than Portal in my opinion. There's even a bonus mission after you complete it where the game takes the piss out of itself, which is something I wish more games would do.

Anachronox by far has made me laugh the most out of any game I've ever played. It's such a shame that more people don't know about it, as it has some fantastic writing and design. One of these days I'll have to go back and re-play it.

Saint's Row 2 is probably on the top of my list but...hmm lemme think...

Conker's Bad Fur Day/ Live and Reloaded, Luigi's Mansion, Psychonaughts, Thousand Year Door, Portal, Portal 2, Fallout 3, New Vegas, Destroy All Humans, Earthworm Jim, Road Rash, Wind Waker, Final Fantasy IX, Saint's Row, Saint's Row 3...that's about it I think. I don't remember enough about Gex, I loved Duke Nukem 3D but didn't really play it for humor and, Celebrity Deathmatch was a steaming pile of shit. Also I would have mentioned Duke Nukem Four? Never! but you couldn't kill enemies with poo! You couldn't even get FRIENDLY kills with poo and the humor truly died when Duke made the rape-joke.

That stuff above is in no particular order by the way. I laughed a lot more at Psychonaughts than I did at Luigi's Mansion.

There are a lot of games that made me laugh, but nothing did it as well as Gears of War 2 did.

No, seriously. Try playing the game as a satire. I know it's most definitely not a satire, but still, just use your imagination. Both me and my co-op partner laughed like all hell throughout the entire game, because if you pretend that the game actually isn't taking itself seriously, it's... just hilarious.
