Personally, I was really gripped by Yoshimo's betrayal in Baldur's Gate 2. I know, I know... A very old, widely respected game, so perhaps a bit of a boring answer. However, having played games from a very young age (from the very first PoP with my grand-father on his 368 back in the late 80's), and an advent sampler of many genres, I have never been quite as gripped by a synthetic character's actions as I was then.

I guess the reason why this moment hit home with me was the result of the following

Character's in games have to be 'functional' besides well developed, as is the case in film and even more so in literature. Take Mordin Solus (the much loved salarian scientist from Mass Effect 2, for those not in the know) for instance; a very funny, witty and quirky character, with a well developed back-ground story that unfolds before players eyes as they progress through the narrative of the game. However, the mere fact that Dr. Solus is a great addition to the game's story, being an action-oriented (note the careful labeling of the game, ME2 fans get a bit nuts about the game's genre...) game, Mordin would have a rubbish character overall, for he would not have been applicable in the combat sequences.

Another example would be Aeris from FFXII, the only (if I remember this correctly) proper healer in the entire party, dies before the conclusive boss-fight.

!If you have not played the game yet, and you plan to, SPOILER ALERT!

Now, Yoshimo shows up very early in the game as the third optional (the player HAS to recruit Imohen) party member. He is mysterious, interesting, sarcastic and a bad ass cool mother-forker! (well, he was back in the day of hand-drawn backgrounds and graphics that required the players imagination...) But besides of of this, he was also the only party-member that could wield katana's (one of the game's conundrums was whether to invest skill points in Katana's as they were extremely rare, but very powerful), set traps and have decent dodging abilities.

And then... hoping to find out what is plaguing this lone wolf, he stabs you in the back, leaves your party (irreversibly and regardless of replaying the game and trying different routes) and all this right before the first (old-school difficult) encounter with the boss-man. Ouch... that stung. My English wasn't all that great back then, so I missed out on a lot of the story (went back and replayed it later at 16, the story was so gripping that it inspired me to get my BA/MA in mythology and oral traditions) but I definitely felt betrayed. Don't get me wrong, I'm no masochist, but having "real" emotional reactions towards in-game events, for me personally any way, occurs but very rarely.

So what character, sequence other moments in gaming will you never forget?

...That's a tough one...WAIT! The romance dialog between Female Shepard and Garrus.

Funniest thing I've ever heard come out of a game. Ever. Her smile looked goofier on my character.

World at War (slight spoiler): When I completed the game on veteran after many, many months of playing the campaign, and failing those last couple of missions over and over again. The best feeling I ever had whilst gaming when the flag was lifted over Berlin, and that achievement popped up on screen.

Left 4 Dead: When I finally got the Zombie Genocidest achievement, on the 4th level of the No Mercy campaign, on top of the hospital in the scaffolding section.

Fallout 3: Killing the Super Mutant which got me to level 30 in The Mall, whilst also getting me the last achievement for 1550G. The best game ever in my opinion.

While not my favorite game, the last level in Braid is by far my favorite moment. That scene constructed the plot that the game had only hinted at and then brilliantly delivered a plot twist purely through gameplay. It was surprising, clever, and a great end to the game. Easily my best single moment in gaming.

Lots of games have great moments, Red dead, Dead Space, God of War, Halo. But by far my favorite is Heavy Rain, if you haven't played the game spoilers obviously.

I have a few favorites, but most recently - League of Legends: Lux triple kill with her Finales Funkeln (shortly followed by an ace). The amount of rage almost ruined the victory,...(*whispers* Not really). If you guys aren't doing at all well in a team push, try not to run away in a straight line down the middle of the lane if there is a competent Lux there. If you do, prepare for devastation.

Edit: I know that's not really a story or moving piece of gaming- but damn it , I was happy about it!

- First entering the city of Vivec in Morrowind. Never before have I seen something this epic in a game.

- The first minutes of Silent Hill 2, when you walk into a deserted town because your dead wife has sent you a letter saying she'd be waiting there. How can this not go wrong?

- The "best" ending of Planescape: Torment. A really touching moment, which I can't say of many in my gaming career.

- Seeing the Ankaran Sarcophagus open in Bloodlines. Also
discovering in hindsight that every word of Rosa's incoherent prophecy actually came true.

The final boss fight from Ocarina of Time. Especially when you're relatively young and new to games, that shit is epic.

Still is, of course.

Undoubtedly, the ending of Shadow of the Colossus.

It's amazing how everything you've been working toward falls apart around you. You're transformed into what is essentially a monster, and then you're defeated by puny little humans (notice what they did there?). The most brilliant part comes as you're being sucked into a mysterious--but clearly deadly--vortex. As you're being pulled across the floor, you can grab onto the stairs to save yourself, but you can't get anywhere. And the grip meter is dropping and your body is flailing and eventually you just...let go. And slip into the vortex and thats that.

I also like that Dormin keeps their promise at the end of it all. And the horse survives, which is nice.

Wow, that was ranty. Still, love that sooooooo much.

Stepping out of the first dungeon in Oblivion, turning to see the Imperial City, and suddenly realizing how huge the game is.

I then went on to spend the next fifty hours playing the game.

SPOLIERS:

Fallout 3: Seeing your dad die, makes the past 10 hours finding him all for nothing.

Red Dead Redemption - The end, damn it John never trust the government.

Call Of Duty 4 - Sniper levels, Macmillian is awesome and the final shootout is tense.

Guitar Hero 2 - Completing Free Bird the first time on medium, I honestly fell to my knees at the end.

I really liked the confrontation with Jedi Council right before the endgame in KOTOR 2. You can only get this is you played a good character but damn, I thought it was a really good scene.

It really solidified my opinion on Kreia being one of the best villains in any game.

Final Fantasy 9 Cinimatic: Assault on the Lifa Tree (49). Inspiring moment that gave me the confidence to write my books. Speaking of which...

Now that I think about it though, Final Fantasy 9 was filled to the brim with fantastically done Cinimatics. That game was one of the last masterpieces of Squinex.

EDIT: If you like watching Boss Battles, feel free to do so. But otherwise, the part I wish for you to watch ends 2:50.

This is so hard... Here's the first two that come to mind.

FFVII: The moment at the end where the lifestream emerges, everyone is blinded by light, and Aeris' eyes open.

Planescape: Torment: The moment when you discover your true name.
