It can be from Half-life, Half-life 2, or any episodes. Mine was when the Niliath is Killed and you choose to work for him in Half-life.

That's easy, the first time you are teleported into another dimension. It's the first time that you truely feel that things are a lot worse than you believed.
I love the fact that Half Life is more the sum of it's parts than any individual spectacular moments.
Probably the bit when Breen tries to run away, and you stop him teleporting away. "Time..Doctor Freeman..Is it really that time again?" Brilliant.
Whenever 'G-Man' speaks is always great.
Half Life: I think the first time you visited the Alien world in Half Life was the most amazing because I really wasn't expecting it (from how the game was up until that point) and I would never have pictured it how it turned out. Although it looks incredibly dated now.
Half Life 2: Ravenholm. It was excellent.
Ep 1: I liked the Strider battle near the end just because of how they move is really well done.
Ep 2: Dog fighting the Strider.

It's all too awesome to say, but i've got to mention the antlion fight in the tunnels. Good bit of fun there.
Throughout the few years before HL2 came out, I was just a wee lad, collecting gameplay and beta videos as much as I could. I remember one video in particular, which was in the prison level, a few combine were behind some beds that were turned on their sides, pathetically trying to destroy my unstoppable bug minions. I dreamed for days just thinking of having my own little minions, sending them to their dooms while laughing hysterically as I watch Combine be thrown from one side of a room to the other. My favorite part was throwing a bugbait and watch every single antlion I had following me be shot down to their doom, and then, just as quickly, another squad came to take its place as cannon fodder in my arsenal. Or, a few steps after you defeat the antlion guard (in the prison), taking out the Combine's shield, then running back to the Warden's room to watch as hundreds of my antlions get torn to pieces by active fire, slowly picking off Combine soldiers one by one until finally the last turret beeps and then shuts down. So much fun.
