Second would be Vamp but he's bisexual...
Then again, I think Purple McHatterson is too... I'm terrible at this...

Off the top of my head (probably forgetting some excellent film characters because my head is stuck in television/books at the moment)...

Overwhelming #1:


Omar Little from The Wire. Not only is he a deep, rich, fully realized and well written character with a tremendous arc, he's about as far from a stereotypically flamboyant homosexual as it's possible to get.

#2:


Kaorin from Azumanga Daioh. Why? Because she's adorable, and tragically hilarious, and because Azumanga Daioh is fucking awesome.

#3:


Wallace Wells from Scott Pilgrim. Not the piece of shit movie, but the exponentially superior series of graphic novels on which it was (very loosely and poorly) based.

#4:


Renly Baratheon. Somewhat loveable, reliably funny character who managed to make all his scenes memorable despite a relatively limited role in the books.

#5:


Dumbledore. Because Dumbledore.

Honorable mention for Tara from BTVS I guess. Willow was not homosexual, she was bisexual.

EDIT: Ahhh shit. And Ennis Del Mar from Brokeback Mountain. Damn it, I KNEW I was forgetting films. There will be more too. Goddam films! Let's just say my 5 are excluding films.

Omar still #1 though regardless.

Since Dumbledore and Renly Baratheon have already been mentioned multiple times, I'll give a shout-out to some as-of-yet-unmentioned characters that I rather like: Ringil and Archeth from the A Land Fit for Heroes book series. They're both fully-realized characters, with real struggles; there's no skirting around the fact that they are homosexual (no "hinting" at their sexualities here, since it's a series that seems to be trying to grab the "grittiest fantasy series" title from A Song of Ice and Fire) while still not making their sexualities their sole defining trait; and - what's really unusual about them - they're two of the three main characters of the series.

Ok, I came here to educate myself more than anything, as, after reading the title, I realised that I can think of very few Homosexual characters, and none that I like enough to consider a favourite. But things worked out, because. . .

Yeah, it's way less than obvious and mostly only hinted at in the story. But here's Ken Levine talking about it some in an interview.

I heard the screenwriter for Fang and Vanille also wrote Asellus' route in Saga Frontier. Hence, as far as I'm concerned the implied (not-implied?) relationship between Fang and Vanille was deliberate.

Kanaya from homestuck.

I mean, I guess there are homosexuals in fiction that I'm more fond of, but Kanaya alone provides a highly unique perspective on the concept of homosexuality. She is a homosexual in a society where bisexuality is the norm.

It's a part of him but it doesn't define him.

I also like how homophobic statements flow over him like water off a duck's back. That's not to say he deserves such things or should be required to do such things but how he handles it deserves respect.
